目的:研究通塞脉片对大鼠缺血后肢侧支循环建立的影响,并探讨其作用机理。方法:将50只大鼠分为5组,40只结扎右下肢肢动脉制作缺血模型,空白对照组、模型对照组以生理盐水灌胃,阳性对照组以复方丹参片灌胃,通塞脉片高、低剂量组分别以不同浓度通塞脉片灌胃。观察各组大鼠双下肢皮肤温度差值,测定促血管内皮生长因子(VEGF),并作大鼠后肢动脉造影。结果:通塞脉片能够改善肢体缺血症状;促进缺血组织VEGF表达,并能促进侧支循环的建立及侧支血管的新生;动脉造影显示各治疗组侧支较模型组明显增多,以通塞脉片高剂量组更为明显。结论:通塞脉片通过促进大鼠缺血肢体的血管新生、建立侧支循环,从而改善肢体缺血对肢体的损伤,其作用机理与提高VEGF有关。
Objective: To study the effect of Tongsaimai Tablet on the establishment of ischemic hind limb collateral circulation in rats, and to explore its mechanism of action. Methods: 50 rats were divided into 5 groups. 40 right limb limbs were ligated to make ischemia model. The blank control group and model control group were given intragastric administration of normal saline. The positive control group was treated with compound Danshen tablets and gavage. The high- and low-dose groups were administered intragastrically with different concentrations of Tongsaimai tablets. The difference of skin temperature between the lower extremities of each group of rats was observed, and vascular endothelial growth factor (VEGF) was measured. The hind-limb arteriography was also performed. Results: Tongsaimai Tablet could improve the symptoms of limb ischemia; promote the expression of VEGF in ischemic tissue, and promote the establishment of collateral circulation and the regeneration of collateral vessels; arterial angiography showed that the collaterals in the treatment groups were significantly increased compared with the model group. Tongsaimai tablets in high-dose group is more obvious. Conclusion: Tongsaimai Tablet can improve the angiogenesis of ischemic limb of rats and establish the collateral circulation, so as to improve limb ischemia by limb ischemia. Its mechanism of action is related to the increase of VEGF.
