目的:探讨人膀胱癌多药耐药细胞凋亡诱导途径。方法:应用免疫细胞化学、原位DNA末端转移酶标记法、相差显微镜及电镜技术,观察BIU-87/ADM细胞表面Fas抗原的表达和抗APO-1单抗对该细胞存活的影响及其特征。结果: Fas抗原在BIU-87/ADM中有高表达,经抗APO-1单抗作用后,BIU-87/ADM细胞存活率明显降低,光镜及电镜下观察到凋亡细胞的形态学改变,原位DNA末端转移酶标记阳性。结论:抗APO-1单抗能与肿瘤细胞表面的Fas抗原结合,触发人膀胱癌多药耐药细胞凋亡。
Objective: To investigate the induction of apoptosis in human bladder cancer multidrug-resistant cells. Methods: The expression of Fas antigen on BIU-87 / ADM cells and the effect of anti-APO-1 McAb on the cell survival and its characteristics were observed by immunocytochemistry, in situ DNA end-transferase labeling, phase contrast microscopy and electron microscopy . RESULTS: Fas antigen was highly expressed in BIU-87 / ADM cells. The viability of BIU-87 / ADM cells was significantly decreased after anti-APO-1 mAb treatment. Morphological changes of apoptotic cells were observed under light microscope and electron microscope , In situ DNA transferase labeling positive. CONCLUSION: Anti-APO-1 mAb can bind to Fas antigen on the surface of tumor cells and trigger the apoptosis of multidrug-resistant human bladder cancer cells.
