目的:建立羊蹄根中大黄酚的HPLC测定方法。方法:外标法,色谱柱:Agilent ZORBAX SC2 C18柱(250mm×4.6 mm,5μm);流动相:甲醇-0.2%磷酸水溶液(65∶35);检测波长:254 nm;流速:1.0 mL/min;进样量:10μL;柱温:25℃。结果:大黄酚在0.85~5.56μg范围内呈良好的线性关系,r=0.9998(n=5),平均回收率为99.2%,RSD为1.6%(n=6)。结论:该方法操作简便、结果准确、分析快速,可作为羊蹄根中大黄酚的定量分析和质量控制方法。
Objective: To establish a HPLC method for the determination of chrysophanol in the root of Rumex. Methods: External standard method, chromatographic column: Agilent ZORBAX SC2 C18 column (250mm×4.6mm, 5μm); mobile phase: methanol-0.2% phosphoric acid aqueous solution (65:35); detection wavelength: 254 nm; flow rate: 1.0 mL/min ; Injection volume: 10 μL; Column temperature: 25°C. Results: The chrysophanol showed a good linear relationship in the range of 0.85~5.56μg, r=0.9998 (n=5). The average recovery was 99.2% and the RSD was 1.6% (n=6). Conclusion: The method is simple, accurate, and rapid. It can be used as a quantitative analysis and quality control method for chrysophanol in roots.
