目的探讨中药红藤合剂灌肠对葡聚糖硫酸钠(DSS)诱导小鼠结肠炎的影响,为中药灌肠的临床应用提供实验依据。方法给昆明小鼠连续饮用5%的葡聚糖硫酸钠13天,造成小鼠结肠炎模型。实验分为对照组(未处理)、模型组(DSS处理)和实验组(造模后红藤合剂灌肠)。病理切片检测小鼠局部肠道病理变化;ELISA法检测局部肠道组织细胞因子分泌情况;WesternBlot法检测肠道组织IκBα、NF-κB表达。结果模型组小鼠疾病活动指数、病理评分均明显高于对照组,局部组织细胞因子IL-12p、TNF-α明显上升,抑炎因子IL-10表达下降,IκBα表达减弱,NF-κB表达上调,且中药治疗组可以有效地缓解临床症状,调节局部肠道组织IκBα、NF-κB细胞因子的表达。结论中药红藤合剂灌肠对DSS诱导的结肠炎小鼠有较好的治疗作用,而这种作用可能是通过对肠道IκBα、NF-κB的表达产生影响,并进而调节局部细胞因子的表达来达成的。
Objective To investigate the effect of Chinese medicine Hongteng Mixture enema on dextran sodium sulfate (DSS)-induced colitis in mice and provide experimental basis for the clinical application of Chinese medicine enema. Methods Kunming mice were continuously given 5% dextran sodium sulfate for 13 days, resulting in a mouse model of colitis. The experiments were divided into control group (untreated), model group (DSS treatment), and experimental group (after modeling Red Rattan mixture enema). Pathological sections were used to detect the local intestinal pathological changes; ELISA was used to detect the secretion of cytokines in local intestinal tissues; Western Blot was used to detect the expression of IκBα and NF-κB in intestinal tissues. Results The disease activity index and pathological scores in the model group were significantly higher than those in the control group. The levels of IL-12p and TNF-α in the tissue were significantly increased, the expression of the anti-inflammatory cytokine IL-10 was decreased, the expression of IκBα was decreased, and the expression of NF-κB was increased. , And Chinese medicine treatment group can effectively relieve clinical symptoms, regulate local intestinal tissue IκBα, NF-κB cytokine expression. Conclusion Chinese medicine Hongteng mixture enema has a better therapeutic effect on mice with DSS-induced colitis, and this effect may be influenced by the expression of IκBα and NF-κB in the intestine, and then regulate the expression of local cytokines.
