目的探讨利用RNA干扰技术在裸鼠体内抑制survivin基因表达并诱导前列腺癌细胞凋亡的作用。方法设计、构建靶向survivin的重组质粒载体并导入裸鼠皮下移植瘤,称量瘤重,计算抑瘤率,应用RT-PCR法检测survivin基因的干涉效率,TUNEL染色法技术检测细胞凋亡效果。结果PBSsi-survivin A和PBSsisurvivin B两种阳性表达载体转染组均具有显著低瘤重和高抑瘤率特点,两组均显著抑制了Survivin mRNA表达,并诱导前列腺癌细胞凋亡。结论RNAi可有效抑制前列腺癌PC-3细胞survivin基因mRNA表达,减缓肿瘤生长,并诱导细胞凋亡。
Objective To investigate the effect of RNA interference on the expression of survivin gene and the apoptosis of prostate cancer cells in nude mice. Methods The recombinant plasmid vector targeting survivin was constructed and subcutaneously transplanted into nude mice. The tumor weight was weighed and the tumor inhibition rate was calculated. The interference efficiency of survivin gene was detected by RT-PCR and the apoptosis effect was detected by TUNEL staining . Results Both PBSsi-survivin A and PBSsisurvivin B transfected groups showed significantly low tumor weight and high inhibitory rate. Both groups significantly inhibited the expression of Survivin mRNA and induced the apoptosis of prostate cancer cells. Conclusion RNAi can effectively inhibit the expression of survivin mRNA in prostate cancer PC-3 cells, slow the tumor growth and induce apoptosis.
