目的探讨过表达和敲除Rab23对人乳腺癌Bcap-37细胞侵袭迁移能力的影响。方法应用Western blot法检测乳腺细胞HBL-100及乳腺癌细胞Bcap-37、MDA-MB-231、MCF-7中Rab23的表达情况。慢病毒转染乳腺癌细胞Bcap-37,筛选出稳定过表达和敲除Rab23的Bcap-37细胞。划痕愈合试验、Transwell实验检测过表达和敲除Rab23后Bcap-37细胞侵袭迁移能力的变化。结果 Western blot法结果显示,Rab23在乳腺和乳腺癌细胞系中均有表达,且Bcap-37细胞表达水平最高,与HBL-100细胞相比,差异有统计学意义(P<0.01);与空载体Bcap-37细胞比较,过表达Rab23的Bcap-37细胞侵袭迁移能力明显增强(P<0.01);敲除Rab23的Bcap-37细胞侵袭迁移能力明显减弱(P<0.01)。结论 Rab23在乳腺癌细胞Bcap-37的侵袭迁移中发挥着重要作用。
Objective To investigate the effect of over-expression and knock-out of Rab23 on invasion and migration of human breast cancer Bcap-37 cells. Methods Western blot was used to detect the expression of Rab23 in breast cancer cells HBL-100 and breast cancer Bcap-37, MDA-MB-231 and MCF-7. Lentiviral transfection of breast cancer cells Bcap-37, screened Bcap-37 cells overexpressing and knocking out Rab23. Scratch healing test and Transwell assay were used to detect the invasion and migration of Bcap-37 cells after overexpression and knockout of Rab23. Results The results of Western blot showed that Rab23 was expressed in both breast and breast cancer cell lines, and the expression level of Bcap-37 cells was the highest. Compared with HBL-100 cells, the difference was statistically significant (P <0.01) The invasion and migration of Bcap-37 cells over-expressing Rab23 were significantly enhanced compared with Bcap-37 cells (P <0.01). The invasion and migration ability of Bcap-37 cells knocked out Rab23 was significantly decreased (P <0.01). Conclusion Rab23 plays an important role in invasion and migration of breast cancer cell Bcap-37.
