目的探究胆石六号汤剂对胆色素结石豚鼠胆囊肿瘤坏死因子-α(TNF-α)及核转录因子(NF-κB)表达的影响。方法将雌性豚鼠40只随机分成3组,空白组10只,模型组15只,实验组15只。空白组除外,其余2组均采用饮食诱导法,建立胆色素结石模型。实验组,灌胃胆石六号汤剂7.7g·kg-1·d-1,其余2组均灌胃用同体积的0.9%氯化钠。30 d后,观察各组豚鼠成石率,用免疫组织化学技术检测胆囊TNF-α及NF-κB蛋白表达和胆囊病理组织的改变情况。结果模型组成石率为90.62%,明显高于空白组26.76%,表明造模成功;实验组成石率为11.00%,明显低于模型组90.62%,可见药物具有抗胆结石的作用。与空白组比较,模型组的TNF-α、NF-κB蛋白表达量明显增加(P<0.01)。与模型组比较,实验组的TNF-α、NF-κB蛋白表达量明显较少(P<0.05)。结论胆石六号汤剂可以明显减少胆色素结石豚鼠的成石率,这可能与药物降低胆色素结石豚鼠胆组织TNF-α、NF-κB蛋白的表达有关。
Objective To investigate the effect of Ganshi Liu decoction on the expression of tumor necrosis factor-α (TNF-α) and nuclear factor kappa B (NF-κB) in gallbladder porcine pig with biliary pigment stones. Methods Forty female guinea pigs were randomly divided into three groups: blank group (n = 10), model group (n = 15) and experimental group (n = 15). Except the blank group, the other two groups adopted the diet induction method to establish the model of bile pigment stones. Experimental group, gavage of gallstone 6 decoction 7.7g · kg-1 · d-1, the remaining two groups were fed with the same volume of 0.9% sodium chloride. After 30 days, the formation rate of guinea pigs in each group was observed. The expression of TNF-α and NF-κB in gallbladder and the pathological changes of gallbladder were detected by immunohistochemistry. Results The stone composing rate of the model was 90.62%, which was significantly higher than that of the blank group (26.76%), indicating that the model was successful. The rate of composing stone was 11.00%, which was obviously lower than that of the model group (90.62%). Compared with the blank group, the expression of TNF-α and NF-κB in the model group increased significantly (P <0.01). Compared with the model group, the expression of TNF-α and NF-κB in the experimental group was significantly less (P <0.05). CONCLUSION Gandhi 6 decoction can significantly reduce the rate of stone formation in guinea pigs with biliary pigmented stones, which may be related to the decrease of the expression of TNF-α and NF-κB in the bile tissue of guinea pigs with bile pigment stones.
