采用35个形态学形状和272个RAPD标记,对贵州12种兰属植物的亲缘关系进行研究。结果表明,引物S 13,S 26及S139扩增的谱带多、清晰、多态性明显,它们单独及结合使用,能有效地将所有供试种类区分开来。尽管这两种标记系统进行亲缘关系分析时,存在一定的差异性,但仍具有很大的同一性,经相关性检测,二者呈极显著相关。形态学形状分析显示,莲瓣兰及春剑与菅草兰的亲缘关系高度同源,这一结果得到RAPD分析的有力支持。因此,莲瓣兰及春剑应为菅草兰的变种,而不应归为新种。
35 morphological and 272 RAPD markers were used to study the genetic relationship of 12 species of orchid in Guizhou. The results showed that the bands amplified by primers S 13, S 26 and S139 were more clear and more polymorphic. They could be used individually and in combination, which could effectively distinguish all tested species. Although there are some differences in the genetic relationship between the two marker systems, they still have a great identity, and the correlation between them is very significant. Morphological analysis showed that the phylogenetic relationships among Cymbidium nivalis and Spring sword were highly homologous with that of Sugarcane, which was strongly supported by RAPD analysis. Therefore, Cymbidium and spring sword should be a variant of Sugarcane, and should not be classified as a new species.
