目的探讨不同剂量亚硒酸钠对氯化镉诱导的雄性SD大鼠肝细胞的凋亡率、c-Myc和P53蛋白表达的影响。方法将65只健康SPF级SD雄性大鼠随机分为13组,分别设对照(生理盐水)组、氯化镉(5.0、10.0、20.0μmol/kg)组及亚硒酸钠(2.5、5.0、10.0μmol/kg)+氯化镉(5.0、10.0、20.0μmol/kg)联合作用组,每组5只。氯化镉采用一次性腹腔注射染毒,亚硒酸钠采用一次性灌胃染毒。染毒后48 h,用流式细胞仪检测细胞凋亡率,免疫组化法测定c-Myc和P53蛋白的表达量。结果与相同剂量氯化镉组相比,亚硒酸钠剂量为2.5μmol/kg时,全部联合作用组的大鼠肝细胞凋亡率、c-Myc和P53蛋白表达均降低(P<0.05或P<0.01);而亚硒酸钠剂量为5.0、10.0μmol/kg时,部分联合作用组的大鼠肝细胞凋亡率、c-Myc和P53蛋白表达降低(P<0.05或P<0.01)。大鼠细胞凋亡率与c-Myc、P53蛋白表达量均呈正相关(P<0.01)。结论2.5μmol/kg的亚硒酸钠对氯化镉所诱导的大鼠肝细胞的凋亡及c-Myc和P53蛋白表达的升高具有一定的拮抗作用。
Objective To investigate the effects of different doses of sodium selenite on the apoptosis rate of hepatocytes and the expressions of c-Myc and P53 in cadmium chloride-induced male SD rats. Methods Sixty-five male Sprague-Dawley (SD) SD male rats were randomly divided into 13 groups: control (saline) group, cadmium chloride (5.0, 10.0 and 20.0μmol / kg) groups and sodium selenite (2.5, 10.0μmol / kg) + cadmium chloride (5.0,10.0,20.0μmol / kg) in combination group, 5 rats in each group. Cadmium chloride using a single intraperitoneal injection, sodium selenite with a one-time intragastric administration. 48 h after exposure, the apoptosis rate was detected by flow cytometry and the expression of c-Myc and P53 protein by immunohistochemistry. Results Compared with the same dose of cadmium chloride group, when the dose of sodium selenite was 2.5 μmol / kg, the apoptosis rate of hepatocytes and the expression of c-Myc and P53 were decreased in all combined groups (P <0.05 or P <0.01). When the dose of sodium selenite was 5.0 and 10.0 μmol / kg, the apoptosis rate of hepatocytes and the expression of c-Myc and P53 decreased in some combination groups (P <0.05 or P <0.01) . There was a positive correlation between the apoptosis rate of rat and the expression of c-Myc and P53 protein (P <0.01). Conclusion 2.5μmol / kg sodium selenite can antagonize the apoptosis of rat hepatocytes induced by cadmium chloride and the increase of c-Myc and P53 protein.
