神经细胞显著的丝氨酸 /精氨酸富集蛋白 1(NSSR 1)是近期发现的可能的神经细胞特异的前体mRNA剪接蛋白。为了进一步对此基因进行研究 ,制备其特异的抗血清是必需环节。本文在通过电脑软件分析找出其特异片段序列的基础上进行多肽片段合成 ,和匙孔 虫戚血蓝蛋白结合后作为抗原免疫家兔 ,使之产生特异性的抗血清。运用所获抗血清 ,对小鼠多种组织中NSSR 1的表达分布进行了Westernblot分析。并且 ,对小鼠脑组织切片也进行了免疫组织化学分析 ,观察NSSR 1在神经系统细胞内的表达分布。Westerblot分析检测到的蛋白条带的分子量大小与推测的NSSR 1蛋白分子量相似 ,表明是NSSR 1的特异条带。NSSR 1蛋白的表达只存在于大脑和小脑 ,而不存在于肝、肺和肾中。免疫组织化学分析的结果表明NSSR 1蛋白的表达主要分布于神经细胞的细胞核 ,而在细胞质中表达水平较低。
Neuronal-prominent serine / arginine-rich protein 1 (NSSR 1) is a recently discovered neuronal-specific precursor splicing protein. In order to further study this gene, preparation of its specific antisera is an essential part. Based on the analysis of computer software to find out its specific fragment sequence based on the peptide fragment synthesis, and the keyhole limpet hemocyanin protein as antigen immunized rabbits to produce specific antiserum. Using the obtained antiserum, Western blot analysis was performed on the distribution of NSSR 1 in various tissues of mice. In addition, immunohistochemistry was also performed on sections of mouse brain to observe the distribution of NSSR 1 in the nervous system cells. The molecular weight of the protein band detected by Westerblot analysis is similar to that of the predicted NSSR1 protein, indicating that it is a specific band of NSSR1. The expression of NSSR1 protein exists only in the brain and cerebellum, but not in the liver, lungs, and kidneys. The results of immunohistochemical analysis showed that the expression of NSSR1 protein was mainly distributed in the nucleus of neurons but lower in the cytoplasm.
