目的通过建立高血压脑出血小鼠模型,探讨血管平滑肌细胞的凋亡对高血压脑出血的作用,为预防高血压脑出血提供线索。方法选用八月龄的C57BL/6雄性小鼠,利用血管紧张素Ⅱ和L-NAME(N-硝基-L-精氨酸甲酯)构建高血压脑出血小鼠模型,小鼠处死立即取大脑组织并用HE染色进行脑出血的鉴定,分别对脑出血组和对照组的小鼠大脑进行TUNEL染色,计数血管平滑肌细胞凋亡比。结果高血压脑出血大脑血管平滑肌细胞凋亡百分比高于正常组脑血管平滑肌(6.82%vs.2.70%,P=0.002),两者比较具有统计学意义。结论血管平滑肌细胞的凋亡可能是高血压脑出血中的重要要病理改变之一。
Objective To establish a mouse model of hypertensive intracerebral hemorrhage to investigate the effect of apoptosis of vascular smooth muscle cells on hypertensive intracerebral hemorrhage and provide clues for the prevention of hypertensive intracerebral hemorrhage. Methods Eight-month old C57BL / 6 male mice were used to construct hypertensive intracerebral hemorrhage mouse model by using angiotensin Ⅱ and L-NAME (N-nitro-L-arginine methyl ester) Brain tissue was identified by HE staining for cerebral hemorrhage. TUNEL staining was performed on the cerebral brains of the cerebral hemorrhage and control groups respectively, and the ratio of apoptosis of vascular smooth muscle cells was counted. Results The percentage of apoptosis of cerebral vascular smooth muscle cells in hypertensive intracerebral hemorrhage was significantly higher than that in normal group (6.82% vs 2.70%, P = 0.002), both of which were statistically significant. Conclusion The apoptosis of vascular smooth muscle cells may be one of the important pathological changes in hypertensive intracerebral hemorrhage.
