目的观察糖尿病神经性疼痛(DNP)大鼠脊髓星形胶质细胞的变化。方法 SD大鼠30只,分为正常对照(NC)组10只和糖尿病(DM)组20只。DM组腹腔单次注射STZ(80mg/kg),NC组腹腔注射生理盐水。两周后眼眶内眦取血测定血糖值,血糖>16.7mmol/L大鼠作为糖尿病模型。4周后测定大鼠热板反应潜伏时间作为痛阈指标,选取DM组痛阈最低的10只作为DNP模型组。取DNP组大鼠脊髓腰段组织切片。神经胶质纤维酸性蛋白(GFAP)标记星形胶质细胞,观察星形胶质细胞在脊髓背角的分布;采用多媒体彩色病理图像分析系统检测阳性面积(Area)μm2和灰度值(Gray Scale)。结果 DM组痛阈(18.41±6.25)较NC组(37.40±7.35)降低(P<0.01),DNP组GFAP染色阳性脊髓星形胶质细胞面积(1377.37±271.83)μm2和灰度值(8638.21±1861.90)水平较NC组升高(P<0.01)。结论糖尿病神经性疼痛时大鼠脊髓星形胶质细胞被激活增生,表达水平升高,抑制脊髓星形胶质细胞激活可能成为治疗DNP的新策略。
Objective To observe the changes of spinal cord astrocytes in diabetic neuropathic pain (DNP) rats. Methods Thirty SD rats were divided into 10 normal control (NC) groups and 20 diabetic (DM) groups. The rats in DM group were injected intraperitoneally with STZ (80mg / kg), and the rats in NC group were injected intraperitoneally with normal saline. Two weeks later, blood samples were taken from the orbital wall to determine the blood glucose level. Blood glucose> 16.7 mmol / L was used as a model of diabetes. After 4 weeks, the latency of hotplate reaction in rats was determined as the threshold of pain threshold. Ten mice with the lowest pain threshold in DM group were selected as DNP model group. Tissue sections of spinal cord of DNP group were taken. Glial fibrillary acidic protein (GFAP) was used to label astrocytes to observe the distribution of astrocytes in the dorsal horn of spinal cord. The colorimetric system was used to detect the positive area (Area) μm2 and Gray Scale ). Results The pain threshold (18.41 ± 6.25) in DM group was significantly lower than that in NC group (37.40 ± 7.35) (P <0.01). The area of astrocytes with GFAP staining in DNP group was 1377.37 ± 271.83 μm 2 and the gray value was 8638.21 ± 1861.90) than NC group (P <0.01). Conclusions In diabetic neuropathic pain, astrocytes in rat spinal cord are activated and proliferated, the expression level is increased. Inhibition of spinal cord astrocyte activation may be a new strategy for the treatment of DNP.
