目的:应用超高效液相色谱-四级杆串联飞行时间质谱法(UPLC-Q-TOF-MS)对江西虫草提取物的主要化学成分进行定性研究。方法:采用Agilent SB-C_(18)(50 mm×4.6 mm,1.8μm)色谱柱,以0.1%甲酸水(A)-乙腈(B)溶液为流动相,梯度洗脱,流速为0.25 mL/min,柱温为30℃;质谱分析采用信息关联模式(IDA)正、负离子分别采集。结果:应用Peak View软件的Formula Finder等功能以及各色谱峰的质谱数据与数据库匹配结合各色谱峰的二级碎片裂解规律,从江西虫草中共鉴定出29个化合物,其中12个化学成分为虫草中首次报道;其主要化学成分包括生物碱、甾醇、糖苷、核苷、氨基酸等。结论:该方法精确、可靠、高效,适用于江西虫草成分的快速鉴定,为江西虫草的开发利用以及阐明其药效物质基础提供参考。
OBJECTIVE: To study the chemical constituents of Cordyceps militaris in Jiangxi by UPLC-Q-TOF-MS. METHODS: The mobile phase was eluted with a gradient of 0.1% formic acid in water (A) -acetonitrile (B) using a Agilent SB-C 18 (50 mm × 4.6 mm, 1.8 μm) column at a flow rate of 0.25 mL / min, and the column temperature was 30 ℃. Mass spectrometry was used to collect positive and negative ions using the information-related mode (IDA). Results: Using Peak View software such as Formula Finder and other chromatographic peaks and mass spectrometry data matching database combined with the chromatographic peak secondary fragmentation law, from Jiangxi Cordyceps identified a total of 29 compounds, of which 12 chemical constituents of Cordyceps For the first time reported; its main chemical components include alkaloids, sterols, glycosides, nucleosides, amino acids and so on. Conclusion: The method is accurate, reliable and efficient and can be applied to the rapid identification of Cordyceps components in Jiangxi Province. It provides a reference for the development and utilization of Cordyceps militaris in Jiangxi Province and the elucidation of the material basis of its efficacy.
