目的:探讨SLC5A8基因甲基化在贲门癌变过程中的作用。方法:采用甲基化特异性PCR法检测42例贲门腺癌及15例癌旁组织SLC5A8基因的甲基化,采用RT-PCR方法检测42例贲门癌组织SLC5A8基因表达水平。结果:42例GCA组织中有18例(43%)SLC5A8基因甲基化阳性。15例癌旁贲门组织中有2例(13%)甲基化阳性。癌组织的甲基化阳性率(43%)显著高于癌旁正常组织(13%)(P<0.01)。贲门癌组织SLC5A8基因甲基化状况与肿瘤分化程度,肿瘤体积,淋巴结转移情况和远处转移情况等临床病理因素均无相关性。18例SLC5A8基因甲基化阳性标本中仅有3例(17%)检测到SLC5A8基因的表达;而24例SLC5A8基因甲基化阴性标本中有10例(42%)检测到SLC5A8基因的表达;SLC5A8基因甲基化阳性的贲门癌组织中SLC5A8基因表达率显著低于甲基化阴性的贲门癌组织的发生率(P<0.05)。结论:SLC5A8基因甲基化是GCA癌变过程中的重要分子事件,是SLC5A8基因失活的重要机制之一。
Objective: To investigate the role of methylation of SLC5A8 gene in carcinogenesis of cardiac cancer. METHODS: Methylation-specific PCR was used to detect the methylation of SLC5A8 gene in 42 cases of gastric adenocarcinoma and 15 paracancerous tissues. RT-PCR was used to detect the expression of SLC5A8 gene in 42 cases of cardiac cancer. RESULTS: Methylation of SLC5A8 was positive in 18 of 42 GCA tissues (43%). Twenty-five cases (13%) of the cancer adjacent to cardiac tissue were positive for methylation. The positive rate of methylation in cancer tissues (43%) was significantly higher than that in normal tissues (13%) (P<0.01). The methylation status of SLC5A8 gene in cardiac cancer tissues was not correlated with the clinical pathological factors such as tumor differentiation, tumor volume, lymph node metastasis and distant metastasis. SLC5A8 gene expression was detected in only 3 (17%) of the 18 SLC5A8 gene methylation-positive samples, and SLC5A8 gene expression was detected in 10 (42%) of the 24 SLC5A8 gene methylation-negative samples. The SLC5A8 gene expression rate in methylation-positive cardiac cancer tissues of SLC5A8 gene was significantly lower than that in methylation-negative cardiac cancer tissues (P<0.05). Conclusion: The methylation of SLC5A8 gene is an important molecular event in the carcinogenesis of GCA and one of the important mechanisms of the inactivation of SLC5A8 gene.
