参照周光宇提出外源DNA直接导入植物组织的原理,改进了某些技术方法,将黑米稻DNA 导入高产品种,取得一定效果。我们采用离体稻恒温水浴法促使开花同步化试验,结果表明白米品种在40.5℃水温催花7—9分钟,黑米品种在29℃水温催花7—8分钟开花同步效果较好。为预防花柱切口在高温多湿条件下的霉变,提高结实率和种子发芽率,进行了 EDTA、苯甲酸钠、CaCl_2等缓冲液的筛选,初步认为用0.2%CaCl_2作切口预处理效果较好。通过 DNA 分子直接导入的方法,已育成了产量高、糙米蛋白也较高的新品系——黑优粘96。
With reference to the principle that Zhou Guangyu directly introduced foreign DNA into plant tissues, some technical methods have been improved to introduce black rice DNA into high-yielding varieties and achieve certain effects. We use the in vitro rice thermostatic water bath method to promote the flowering synchronization test, the results showed that white rice species in the water temperature of 40.5 ℃ spent 7-9 minutes, black rice varieties in the 29 ℃ water temperature for 7-8 minutes to flowering synchronization better. In order to prevent mildew, increase seed setting rate and seed germination rate of style cut under high temperature and humidity conditions, screening of buffer such as EDTA, sodium benzoate and CaCl 2 was carried out. It was preliminarily believed that 0.2% CaCl 2 as an incision pretreatment was better. Through the direct introduction of DNA molecules, a new line with high yield and high brown rice protein has been bred.
