目的:建立鄂西北地区白及化学成分的HPLC指纹特征图谱,为白及药材的质量控制提供科学依据。方法:采用HPLC法,以Waters SunFire C18(4.6 mm×250 mm,5μm)为色谱柱,Diamonsil C18保护柱,以乙腈和水为流动相,梯度洗脱,流速为1 ml·min-1,检测波长270 nm,柱温为30℃。结果:不同产地的15批白及药材化学成分特征图谱共检出9个分离度良好的共有峰。对不同产地的9个特征峰进行聚类分析,白及药材产地与特征图谱相似度差异不大,样品中参照物1,4-二[4-(葡萄糖氧)苄基]-2-异丁基苹果酸酯(militarine)的含量有明显差别。结论:该方法简便、快速,可做为白及的质量控制的有效手段。
Objective: To establish HPLC fingerprints of white and chemical constituents in the northwestern Hubei Province, and to provide a scientific basis for quality control of white and medicinal materials. Methods: The HPLC method was used on a Waters SunFire C18 (4.6 mm × 250 mm, 5 μm) column and a Diamonsil C 18 guard column. The mobile phase consisted of acetonitrile and water with a gradient of 1 ml · min -1. Wavelength 270 nm, column temperature 30 ℃. Results: The characteristic peaks of chemical composition of 15 batches of white medicinal materials in different habitats were detected. Nine common peaks with good separation were detected. The cluster analysis of nine characteristic peaks in different habitats showed that there was no significant difference in similarity between the origin and the characteristic map of white and medicinal materials. The reference substance 1,4-bis [4- (glucoseoxy) benzyl] -2-isobutyl There is a clear difference in the content of militarine. Conclusion: The method is simple, rapid, and can be used as an effective means of quality control.
