本试验研究了色氨酸合成酶合成2-L-甲基色氨酸,验证色氨酸合成酶基因工程菌活性,用单因素实验及响应面分析法对色氨酸合成酶合成2-L-甲基色氨酸的合成条件进行优化。结果表明,最佳的合成条件:pH=8.06,温度为33.3℃,L-丝氨酸浓度为12.7 g/L,此条件下,手性药物2-L-甲基色氨酸的合成量为10.5 g/L。研究结果可为手性药物2-L-甲基色氨酸工业化生产提供参考价值。
In this study, tryptophan synthetase synthesis of 2-L-methyl tryptophan, tryptophan synthase genetically engineered bacteria activity verified by single factor experiments and response surface analysis of tryptophan synthase synthesis of 2-L - methyl tryptophan synthesis conditions were optimized. The results showed that the optimal synthesis conditions were as follows: pH = 8.06, temperature 33.3 ℃, L-serine concentration 12.7 g / L. Under these conditions, the amount of synthesized 2-L-methyl tryptophan was 10.5 g / L. The results provide a reference value for the industrial production of 2-L-methyltryptophan, a chiral drug.
