目的:分析研究黄芪多糖对异丙肾上腺素诱导的乳鼠心肌细胞肥大的保护作用。方法:分离乳鼠心肌细胞进行原代培养,以异丙肾上腺素诱导,构造乳鼠心肌细胞肥大模型,分为5组,分别为空白对照组、黄芪多糖高、中、低剂量组和阳性对照组,以黄芪多糖处理,培养后进行心肌细胞蛋白质含量的测定和心肌细胞体积的测定,分析黄芪多糖对异丙肾上腺素诱导的乳鼠心肌细胞肥大的保护作用。结果:经Iso处理可成功诱导乳鼠心肌细胞肥大;高、中、低剂量组比较来看,低剂量组与阳性对照组最为接近,低剂量浓度抑制效果更佳,即黄芪多糖0.1 mg/L能有效对抗心肌的肥厚性改变。结论:黄芪多糖对异丙肾上腺素诱导的肥大心肌细胞具有较好的保护作用,其机制可能与抑制TLR4活化、阻断NF-KB及Ca MKⅡ信号通路的激活,减少炎症反应有关。
Objective: To study the protective effect of astragalus polysaccharides on isoprenaline-induced cardiomyocyte hypertrophy in neonatal rats. Methods: Primary cultured neonatal rat cardiomyocytes were isolated and induced by isoprenaline. Hypertrophy models of neonatal rat cardiomyocytes were established and divided into 5 groups: blank control group, high, medium and low dose APS groups and positive control group Group, treated with astragalus polysaccharides. After cultured, the protein content of cardiomyocytes and the volume of cardiomyocytes were measured. The protective effect of astragalus polysaccharides on isoprenaline-induced cardiomyocyte hypertrophy was analyzed. Results: Iso-induced cardiomyocyte hypertrophy can be successfully induced in neonatal rats. Compared with the positive control group, the low-dose group and the positive control group showed the best inhibitory effect, namely, APS 0.1 mg / L Can effectively fight the hypertrophy of the myocardium. CONCLUSION: Astragalus polysaccharide has a good protective effect on isoprenaline-induced hypertrophic cardiomyocytes. The mechanism may be related to the inhibition of TLR4 activation, the activation of NF-κB and Ca-MKⅡ signaling pathway and the reduction of inflammatory reaction.
