分析了小花玄参化学成分,并测定了总黄酮含量。采集自然野生比较成熟的小花玄参,进行样品处理,优化提取工艺,选择连续回流法提取,进行针对溶剂类型、浓度、回流时间、回流次数、水温五个变动参数正交试验,最终以70%甲醇为溶剂,回流时间120min,回流2次,提取温度80℃作为总黄酮检测条件,采用乙醇提取液作为强心苷的监测,石油醚提取液用于甾体类、三萜类物质的检测。对照品浓度c与吸光度A存在线性关系,A=2.264c-0.025,r=0.984,线性范围0.020~0.15mg/ml,相对标准差=0.251,RSD=0.271,最大波长510nm,总黄酮含量(5.625±0.251)%,精密度、稳定性相对保留时间RSD(%)分别为2.46%、1.16%,回收率99.99%。
The chemical constituents of Scrophulariaceae were analyzed, and the content of total flavonoids was determined. The natural and wildly mature Scrophulariaceae Scrophulariaceae were collected to conduct sample processing, optimize the extraction process, and selected for continuous reflux extraction. Five orthogonal parameters for solvent type, concentration, reflux time, number of reflux, and water temperature were orthogonally tested, and finally 70%. Methanol is a solvent, refluxing time 120min, refluxing 2 times, extraction temperature 80°C as the detection condition of total flavonoids, ethanol extract as monitoring of cardiac glycosides, petroleum ether extracts used for the detection of steroids and triterpene substances. There was a linear relationship between reference concentration c and absorbance A, A=2.264c-0.025, r=0.984, linear range 0.020 to 0.15 mg/ml, relative standard deviation=0.251, RSD=0.271, maximum wavelength 510 nm, total flavonoid content (5.625) ±0.251)%, precision, stability relative retention time RSD (%) were 2.46%, 1.16%, and the recovery rate was 99.99%.
