目的了解重组人白细胞介素3(rhIL-3)和重组人白细胞介素6(rhIL-6)对骨髓造血微环境的调节作用。方法应用成纤维细胞体外集落形成法,观察了rhIL-3和rhIL-6对成纤维细胞集落(CFU-F)体外生长的影响。结果rhIL-3和rhIL-6能显著地刺激CFU-F的生长,两者联合应用时对CFU-F生长的刺激作用明显强于两者单用,表现出协同效应。结论rhIL-3和rhIL-6可促进基质细胞生长,对促进化疗和(或)放疗及骨髓移植后造血微环境的恢复和重建具有重要的临床应用潜能
Objective To investigate the effect of recombinant human interleukin 3 (rhIL-3) and recombinant human interleukin 6 (rhIL-6) on hematopoietic microenvironment. Methods In vitro fibroblasts colony formation assay was used to observe the effects of rhIL-3 and rhIL-6 on fibroblast colony (CFU-F) growth in vitro. Results rhIL-3 and rhIL-6 could significantly stimulate the growth of CFU-F. The combination of rhIL-3 and rhIL-6 had a stronger stimulatory effect on the growth of CFU-F than that of the two alone, showing a synergistic effect. Conclusion rhIL-3 and rhIL-6 can promote the growth of stromal cells, and have important clinical potential for promoting the recovery and reconstruction of hematopoietic microenvironment after chemotherapy and/or radiotherapy and bone marrow transplantation.
