目的观察弓形虫可溶性速殖子抗原(Soluble tachyzoite antigen,STAg)与BCG-DNA和A(lOH)3佐剂联合免疫小鼠的免疫效果。方法将BALB/c小鼠随机分为5组,分别经小鼠头背部皮下注射20μg STAg、50μg BCG-DNA+20μgSTAg、50μg A(lOH)3+20μg STAg、50μg BCG-DNA+50μg A(lOH)3+20μg STAg,对照组注射生理盐水,共免疫4次。末次免疫后1周处死小鼠,采血分离血清,并制备脾细胞悬液,ELISA法测定小鼠血清IgG值及抗体效价;ELISPOT法检测分泌特异性IFNγ的淋巴细胞数量。结果 A(lOH)3+BCG-DNA+STAg组小鼠血清抗体效价和IgG值及产生特异性IFNγ的淋巴细胞数量均显著高于STAg组、BCG-DNA+STAg组和A(lOH)3+STAg组(P<0.05)。结论 STAg联合BCG-DNA和A(lOH)3佐剂能诱导小鼠产生更强的体液免疫和细胞免疫应答,A(lOH)3与BCG-DNA佐剂对弓形虫STAg具有免疫协同效应。
Objective To observe the immune effect of Tachyzoite soluble tachyzoite antigen (STAg) combined with BCG-DNA and A (lOH) 3 adjuvant in mice. Methods BALB / c mice were randomly divided into 5 groups. The mice were injected subcutaneously with 20μg STAg, 50μg BCG-DNA + 20μgSTAg, 50μg A (lOH) 3 + 20μg STAg, 50μg BCG-DNA + ) 3 + 20μg STAg, the control group was injected with saline and co-immunized 4 times. The mice were sacrificed one week after the last immunization, blood was collected to separate the serum, and splenocyte suspension was prepared. The serum IgG value and antibody titer were measured by ELISA. The number of lymphocytes secreting specific IFNγ was measured by ELISPOT. Results The titer and IgG value of serum antibody and the number of lymphocytes producing specific IFNγ in A (lOH) 3 + BCG-DNA + STAg group were significantly higher than those in STAg group, BCG-DNA + STAg group and A (lOH) 3 + STAg group (P <0.05). Conclusion STAg combined with BCG-DNA and A (lOH) 3 adjuvant can induce stronger humoral and cellular immune responses in mice. A (lOH) 3 and BCG-DNA adjuvant have an immunocompetent effect on STAg.
