应用流式细胞光度分析法测定丹参和川芎嗪对体外培养的瘢痕成纤维细胞DNA相对含量和细胞周期时相变化,发现药物在一定浓度作用下,DNA指数无明显变化;丹参使C2M期细胞分布增多,G2M期延长;川芎嗪使G2M期细胞分布增多,G2M期、S期延长;药物使细胞群体倍增时间延长,与浓度呈明显直线正相关。认为,丹参能抑制细胞的有丝分裂,川芎嗪能抑制细胞的DNA合成、复制和有丝分裂,细胞周期均停滞于G2M期
The flow cytometric assay was used to determine the DNA content and cell cycle changes of S. miltiorrhiza fibroblasts in vitro cultured with S. miltiorrhiza and Tetramethylpyrazine. It was found that under certain concentration of drugs, the DNA index had no significant change; S. miltiorrhiza make C2 M phase The cell distribution increased and the G2M phase prolonged. Ligustrazine increased the G2M phase cell distribution, G2M phase and S phase prolonged; the drug extended the cell population doubling time and showed a positive linear correlation with the concentration. It is believed that salvia miltiorrhiza can inhibit cell mitosis, tetramethylpyrazine can inhibit DNA synthesis, replication, and mitosis, cell cycle arrested in G2-M phase
