目的研究苦参碱(Mat)对体外培养的Hela细胞增殖细胞核抗原(PCNA)表达及细胞周期的影响,为Mat治疗类风湿关节炎(RA)提供可能的实验室依据。方法用免疫组织化学法测定PCNA表达水平,用流式细胞术测定细胞周期变化,并与甲氨蝶呤(MTX)作比较。结果Mat、MTX均可使体外培养的Hela细胞的PCNA表达水平明显降低,还可以使细胞周期发生明显的变化,干预组Hela细胞停留在G1期的比例要比未干预组明显增加(P<0.05),而S、G2期的细胞比例比未干预组则明显减少(P<0.05)。结论苦参碱有下调Hela细胞PCNA表达水平,阻滞细胞周期的作用,且机制与MTX有相似之处。推测Mat有治疗RA的前景。
Objective To study the effects of matrine on the expression of proliferating cell nuclear antigen (PCNA) and cell cycle in Hela cells cultured in vitro, and to provide a possible laboratory basis for the treatment of rheumatoid arthritis (RA) by Mat. Methods The expression of PCNA was determined by immunohistochemical method. Cell cycle changes were determined by flow cytometry and compared with methotrexate (MTX). Results Both Mat and MTX could significantly reduce the expression of PCNA in Hela cells cultured in vitro and also significantly change the cell cycle. The proportion of Hela cells in the G1 phase in the intervention group was significantly higher than that in the non-intervention group (P<0.05). The proportion of S and G2 cells was significantly lower than that of the non-intervention group (P<0.05). Conclusion Matrine can down-regulate the expression of PCNA in Hela cells and block cell cycle, and the mechanism is similar to that of MTX. It is speculated that Mat has the prospect of treating RA.
