目的 :观察重组葡激酶 (r SAK)在不同剂量下凝血功能指标的变化规律。方法 :将 2 0只体重为 2 .0~ 2 .5kg新西兰兔 (雄性 )使其兔球囊损伤致兔股动脉血栓形成 ,并随机分成 4组 ,第一组为对照组 ,注生理盐水 10ml;第二、三、四组为试验组 ,分别注r SAK 1.5× 10 4u/kg、3.0× 10 4u/kg、6 .0× 10 4u/kg ,各组分别在不同时间进行采样。结果 :经r SAK不同剂量溶栓后纤维蛋白原(FIB)均有显著下降 ,第三、四组溶栓后 12 0min凝血酶原时间 (PT)延长 ,而活性部分凝血激酶时间 (APTT)、凝血酶时间 (TT)均无显著延长 ;血小板 (Plt)各组在球囊损伤后均显著降低 ,但试验组在溶栓前、溶栓后不同时间内与对照组比较无显著性差异。结论 :球囊损伤致兔股动脉血栓形成 ,r SAK在有效溶栓剂量范围内 ,对凝血功能无显著影响 ,是一种高效且安全的溶栓药物
Objective: To observe the changes of coagulation function of recombinant staphylokinase (r SAK) under different doses. Methods: 20 rabbits (male) weighing 2.5-2.5 kg were inflicted with rabbit balloon femoral artery thrombosis caused by balloon injury and were randomly divided into 4 groups. The first group was control group, saline 10ml ; The second, third and fourth groups were the experimental group, respectively, injected with 1.5 × 10 4u / kg SA, 3.0 × 10 4u / kg and 6.0 × 10 4u / kg groups, respectively, at different times for sampling. Results: The fibrinogen (FIB) decreased significantly after thrombolysis with r SAK. The thromboplastin time (PT) at 120 minutes after the thrombolysis in the third and fourth groups was prolonged, while the activity of active partial thromboplastin time (APTT) Thrombin time (TT) were not significantly prolonged; platelet (Plt) after balloon injury in each group were significantly reduced, but the test group before thrombolysis, thrombolysis at different times compared with the control group no significant difference. CONCLUSIONS: Rabbit femoral artery thrombosis caused by balloon injury has no significant effect on coagulation function in the range of effective thrombolytic dose, and is an efficient and safe thrombolytic drug
