目的:研究VEGFR-3、podoplanin和CD34标记的阳性脉管在宫颈癌组织中表达的特点,分析其与肿瘤临床病理特点之间的关系。方法:采用双标及单标免疫组化方法检测58例宫颈鳞癌组织VEGFR-3、podoplanin和CD34标记阳性脉管的密度。结果:癌灶边缘组织VEGFR-3和podoplanin阳性脉管密度显著高于癌组织(P<0.05),也显著高于对照组宫颈组织(P<0.05);癌灶边缘组织和癌组织CD34阳性脉管密度差异无统计学意义(P>0.05),均高于对照组宫颈组织(P<0.05)。Ⅱa期组癌灶边缘组织VEGFR-3、podoplanin和CD34阳性脉管密度分别为17.46±2.65、14.36±1.21、17.98±4.13显著高于Ⅰb期组的13.52±1.23、10.58±3.25、13.75±3.38(P<0.05);淋巴结转移组癌灶边缘组织VEGFR-3和podoplanin阳性脉管密度18.68±0.89和14.67±3.72显著高于无淋巴结转移组11.45±3.56和10.77±2.65(P<0.05),CD34阳性脉管密度在两组中表达差异无统计学意义(P>0.05)。结论:宫颈癌VEGFR-3、podoplanin和CD34表达与肿瘤临床分期有关,癌灶边缘组织VEGFR-3和podoplanin高表达与淋巴结转移亦密切相关,podoplanin有望成为宫颈癌淋巴结转移预后的一种可靠标志物。
OBJECTIVE: To study the expression of VEGFR-3, podoplanin and CD34 positive vessels in cervical cancer and to analyze their relationship with clinicopathological features. Methods: The densities of VEGFR-3, podoplanin and CD34-labeled positive vasculature in 58 cases of cervical squamous cell carcinoma were detected by double-label and single-label immunohistochemistry. Results: The positive vascular density of VEGFR-3 and podoplanin in the marginal tissue of the lesion was significantly higher than that in the cancerous tissue (P <0.05), and also significantly higher than that in the control group (P <0.05) There was no significant difference in tube density between the two groups (P> 0.05), which were all higher than those in the control group (P <0.05). The positive vascular densities of VEGFR-3, podoplanin and CD34 in the margins of stage IIa were 17.46 ± 2.65, 14.36 ± 1.21 and 17.98 ± 4.13, respectively, which were significantly higher than those of group Ⅰb 13.52 ± 1.23,10.58 ± 3.25,13.75 ± 3.38 P <0.05). The positive vascular densities of VEGFR-3 and podoplanin in the marginal tissue of lymph node metastasis group were significantly higher than those in non-lymph node metastasis group (11.45 ± 3.56 and 10.77 ± 2.65, 18.68 ± 0.89 and 14.67 ± 3.72, respectively) Vascular density in the two groups showed no significant difference (P> 0.05). Conclusion: The expression of VEGFR-3, podoplanin and CD34 in cervical cancer is related to the clinical stage of carcinoma. The high expression of VEGFR-3 and podoplanin in the marginal tissue of cancer is closely related to lymph node metastasis, and podoplanin is expected to become a reliable marker of prognosis of cervical cancer with lymph node metastasis .
