目的:探讨舒血宁注射液对大鼠心肌缺血再灌注损伤的保护作用。方法:将50只雄性SD大鼠随机分为5组:假手术组、模型组、舒血宁注射液高、中、低剂量组。按文献方法除假手术组左冠脉前降支下只穿线不结扎外,另4组均结扎左冠脉前降支,结扎30min后剪断结扎线再灌注60 min。在结扎前5 min,假手术组和模型组分别腹腔注射生理盐水1 mL/kg;舒血宁注射液高、中、低剂量组分别腹腔注射舒血宁注射液2 mL/kg、1 mL/kg、0.5 mL/kg。再灌注结束后,从大鼠颈动脉取血3 mL,离心制备血清,检测CK、LDH活性;取血后各组均立即取出心脏,制成匀浆,检测SOD、GSH-Px活性及MDA含量。结果:与模型组相比,舒血宁注射液高、中、低剂量组CK、LDH活性显著降低(P<0.01);MDA含量显著降低(P<0.01)。结论:舒血宁注射液对大鼠缺血再灌注损伤心肌有保护作用,可能与舒血宁注射液能够提高缺血再灌注大鼠心肌的抗氧化能力有关。
Objective: To investigate the protective effect of Shuxuening injection on myocardial ischemia-reperfusion injury in rats. Methods: Fifty male Sprague-Dawley rats were randomly divided into 5 groups: sham operation group, model group and Shuxuening injection high, medium and low dose groups. According to the literature method, except the sham group, only the left anterior descending coronary artery was ligated without threading, and the other 4 groups were ligated with the descending anterior descending branches of the left coronary artery. The ligature was ligated for 30 minutes and ligated for 60 minutes. Rats in sham operation group and model group were intraperitoneally injected with normal saline (1 mL / kg) 5 min before ligation, Shuxuening injection (2 mL / kg, 1 mL / kg, 0.5 mL / kg. At the end of reperfusion, 3 mL blood was taken from the carotid artery of rats and centrifuged to prepare CK and LDH activities. After taking blood, the hearts were removed immediately and homogenized. The activities of SOD, GSH-Px and the content of MDA . Results: Compared with the model group, the activities of CK and LDH in Shuxuening injection group were significantly decreased (P <0.01) and MDA content was significantly decreased (P <0.01). Conclusion: Shuxuening injection has a protective effect on myocardial ischemia-reperfusion injury in rats, which may be related to Shuxuening injection can improve myocardial antioxidant capacity of ischemia-reperfusion rats.
