目的 :探讨长链非编码RNA BANCR在非小细胞肺癌(non-small cell lung cancer,NSCLC)组织及细胞系中的表达水平,以及BANCR对NSCLC细胞增殖和侵袭的影响。方法:用定量PCR技术检测NSCLC组织及细胞系中BANCR的表达水平;通过转染pc DNA-BANCR上调BANCR的表达水平,并通过q PCR检测转染效率。用Transwell实验检测上调BANCR的水平对SPC-A1细胞和A549细胞增殖和侵袭能力的影响,Western blot检测这2种细胞中上调BANCR的水平对上皮-间质转化(epithelial-mesenchymal transition,EMT)标志物上皮型钙黏蛋白(E-cadherin)和波形蛋白(Vimentin)表达水平的影响。结果 :相比正常肺组织及细胞,在NSCLC组织和细胞中BANCR的表达出现显著下调。MTT实验显示,上调BANCR的表达能降低SPCA1细胞和A549细胞的增殖和侵袭能力。BANCR过表达能通过上调E-cadherin表达并下调Vimentin表达,影响EMT。结论:BANCR可通过影响EMT,抑制NSCLC细胞的增殖和侵袭。
Objective: To investigate the expression of long-chain non-coding RNA BANCR in non-small cell lung cancer (NSCLC) tissues and cell lines and the effect of BANCR on the proliferation and invasion of NSCLC cells. Methods: The expression of BANCR in NSCLC tissues and cell lines was detected by quantitative PCR. The expression of BANCR was up-regulated by pcDNA-BANCR transfection and the transfection efficiency was detected by q-PCR. Transwell assay was used to detect the effect of BANCR up-regulation on the proliferation and invasion ability of SPC-A1 cells and A549 cells. Western blot was used to detect the expression of BANCR in the two epithelial-mesenchymal transition (EMT) markers Effects of E-cadherin and vimentin on the expression of E-cadherin and Vimentin in human. Results: BANCR expression was significantly down-regulated in NSCLC tissues and cells compared to normal lung tissues and cells. MTT experiments showed that upregulation of BANCR expression can reduce the proliferation and invasion ability of SPCA1 cells and A549 cells. BANCR overexpression can affect EMT by up-regulating E-cadherin expression and down-regulating Vimentin expression. Conclusion: BANCR can inhibit the proliferation and invasion of NSCLC cells by affecting EMT.
