目的:探讨碱性成纤维细胞生长因子(basic fibroblast growth factor,FGF-2)对肺癌A459细胞株pim-3的影响。方法:不同浓度的(0、25、50、100ng/ml)FGF-2在体外作用于肺癌A459细胞,半定量逆转录聚合酶链式反应(RT-PCR)法检测pim-3mRNA,Western blot检测pim-3蛋白表达。结果:与空白对照组比较,RT-PCR和Western blot检测结果显示pim-3mRNA及其蛋白的表达在各FGF-2作用组均显著增高,各组间比较差异有统计学意义(P<0.01)。结论:FGF-2可以量效的方式上调pim-3的表达。
Objective: To investigate the effect of basic fibroblast growth factor (FGF-2) on the expression of pim-3 in lung cancer cell line A459. METHODS: FGF-2 at different concentrations (0, 25, 50, 100 ng / ml) was used to effect A459 lung cancer cells in vitro. Pim-3 mRNA was detected by semi-quantitative reverse transcriptase polymerase chain reaction pim-3 protein expression. Results: The results of RT-PCR and Western blot showed that the expression of pim-3 mRNA and its protein were significantly increased in each group of FGF-2 compared with the blank control group, the difference was statistically significant (P <0.01) . CONCLUSION: FGF-2 up-regulates pim-3 expression in a dose-efficient manner.
