通过试验选择聚乙二醇(PEG 1000)与聚乙烯吡咯烷酮(PVP 30000)为组合表面活性剂,(NH4)2SO4-H2O为双水相体系。在VPEG 1000/VPVP 30000=2.00 mL/2.00 mL,(NH4)2SO4用量为1.5 g,pH 6.0的B.R缓冲溶液最佳萃取条件下,经两次萃取,桑叶中芦丁定量进入PEG-PVP相。用紫外分光光度法在375 nm波长处直接萃取测定桑叶提取液中的芦丁含量。试验结果表明,方法的相对标准偏差≤3.8%(n=7),样品加入回收率为96.5%。试验表明,方法具有良好的选择性。此外,用试样超声提取率较常规索氏提取率高。
Polyethylene glycol (PEG 1000) and polyvinylpyrrolidone (PVP 30000) were selected as the combined surfactant by experiments, and (NH4)2SO4-H2O was a two-phase system. Under the optimum extraction conditions of VPEG 1000/VPVP 30000=2.00 mL/2.00 mL, (NH4)2SO4 1.5 g, pH 6.0 BR buffer solution, after two extractions, rutin in mulberry leaves quantitatively enters the PEG-PVP phase. . The content of rutin in the extract of mulberry leaves was determined directly by ultraviolet spectrophotometry at a wavelength of 375 nm. The test results show that the relative standard deviation of the method is ≤3.8% (n=7) and the recovery rate of the sample is 96.5%. Tests show that the method has good selectivity. In addition, the ultrasonic extraction rate of the sample is higher than the conventional Soxhlet extraction rate.
