用c-src基因表达产物pp60 ̄(c-src)的特异性单克隆抗体(McAb327)对35例食管鳞癌及22例癌旁组织进行免疫组化研究。结果:食管鳞癌、癌旁增生及正常鳞状上皮pp60 ̄(c-src)阳性率分别为65.7%(23/35)、100.0%(15/15)、57.1%(4/7),差异有显著性(P<0.05);鳞癌细胞pp60 ̄(c-src)表达量高于癌旁增生及正常鳞状上皮细胞。不同分化程度鳞癌(Ⅰ、Ⅱ、Ⅲ级)pp60 ̄(c-src)阳性率分别为80.0%(8/10),68.4%(13/19),33.3(2/6),差异无显著性(P>0.05);分化程度越高,pp60 ̄(c-src)表达量越高(P<0.005)。结果揭示,c-src基因激活表达与食管鳞癌发生发展有关,在其癌变过程中pp60 ̄(c-src)表达量升高,并与食管鳞癌分化有关。
Thirty-five patients with esophageal squamous cell carcinoma and 22 paracancerous tissues were studied by immunohistochemistry using a monoclonal antibody specific for c-src gene expression product pp60 (c-src) (McAb327). RESULTS: The positive rates of pp60 ̄ (c-src) in esophageal squamous cell carcinoma, adjacent hyperplasia, and normal squamous epithelium were 65.7% (23/35), 100.0% (15/15), and 57.1% ( 4/7), the difference was significant (P <0.05); the expression of pp60 ~ (c-src) in squamous cell carcinoma cells was higher than that in adjacent hyperplasia and normal squamous epithelial cells. The positive rates of pp60 ̄ (c-src) were 80.0% (8/10), 68.4% (13/19), and 33.3 (2/3) of squamous cell carcinomas (I, II, and III), respectively. 6) There was no significant difference between the two groups (P>0.05). The higher the degree of differentiation was, the higher the expression of pp60 (c-src) was (P<0.005). The results revealed that c-src gene activation expression is related to the development of esophageal squamous cell carcinoma. The expression of pp60 ̄ (c-src) is increased in the course of its carcinogenesis and is related to the differentiation of esophageal squamous cell carcinoma.
