目的:探讨红芪多糖(Hedysarum Polybotys sacce,HPS)对链脲佐菌素(Streptozotoein,STZ)诱导的糖尿病模型大鼠视网膜血管内皮细胞生长因子(Vascular endothelial growth factor,VEGF)、色素上皮细胞衍生因子(Pigment epithlium-derived facto,PEDF)和血清C-反应蛋白(C-reactive Protein,CRP)水平的影响及视网膜组织病理学的改变,并对红芪多糖的作用机制进行研究。方法:选取150只健康大鼠,按照随机数字表法分为实验组和对照组,对照组的25只大鼠给予柠檬酸缓冲液腹腔注射,实验组的125只大鼠给予链脲佐菌素腹腔注射。实验组动物模型制备成功后平均分为HPS低剂量组、HPS中剂量组、HPS高剂量组、空白组、多贝斯组,HPS组的大鼠分别给予50mg/kg、100mg/kg、150mg/kg浓度的HPS灌胃,空白组与对照组给予等体积的生理盐水灌胃,多贝斯组给予90mg/kg的多贝斯灌胃,1次/天,连续灌胃8周。最后一次灌胃结束后,取大鼠的血清进行CRP含量检测,同时摘取大鼠的眼球置于甲醛溶液中固定,然后剥取视网膜进行病理切片观察,并检测视网膜组织中VEGF、PEDF的水平。结果:血清CRP检测结果:与对照组相比,空白组大鼠血清CRP含量升高,差异具有统计学意义(P<0.05);与空白组比较,HPS组和多贝斯组血清CRP含量均降低,差异具有统计学意义(P<0.05);与多贝斯组比较,高、中、低浓度HPS组大鼠血清CRP含量降低均有统计学意义(P<0.05);病理切片观察结果:与空白组比较,HPS各组大鼠视网膜受损程度均降低;与多贝斯组比较,HPS高剂量组大鼠的视网膜增厚程度最明显,每一层的细胞排列较整齐。VEGF、PEDF检测结果:与对照组比较,空白组视网膜组织中VEGF水平升高,PEDF含量降低,差异具有统计学意义(P<0.05);与空白组比较,HPS各剂量组和多贝斯组VEGF水平降低,PEDF含量升高,差异具有统计学意义(P<0.05)。结论:红芪多糖能够降低大鼠血清CRP含量,使视网膜组织中VEGF水平降低,PEDF含量升高,遏制视网膜新血管的产生而起到保护视网膜的作用。
Objective: To investigate the effect of Hedysarum Polybotys sacce (HPS) on the expression of retinal vascular endothelial growth factor (VEGF), pigment epithelium-derived factor (VEGF) in diabetic rat model induced by streptozotocin (STZ) (Pigment epithlium-derived facto, PEDF) and serum C-reactive protein (CRP) levels and retinal histopathological changes, and the mechanism of action of Radix Hedysari. Methods: 150 healthy rats were selected and randomly divided into experimental group and control group according to random number table method. 25 rats in control group were given intraperitoneal injection of citrate buffer solution. 125 rats in experimental group were given streptozotocin Intraperitoneal injection. The rats in the experimental group were equally divided into HPS low dose group, HPS middle dose group, HPS high dose group, blank group, Dobesia group and HPS group, and were given 50mg / kg, 100mg / kg, 150mg / kg The rats in Dobesi group were given Dobos at 90mg / kg orally once daily for 8 weeks. At the end of the last gavage, the rat serum was taken for the detection of CRP content. At the same time, the eyes of the rats were removed and fixed in formaldehyde solution. Then the retinas were stripped for pathological observation and the levels of VEGF and PEDF in the retina were detected . Results: Compared with the control group, serum CRP levels in serum of CRP group were significantly increased (P <0.05). Compared with the blank group, serum CRP level of HPS group and Dobbes group were decreased , The difference was statistically significant (P <0.05); Compared with the dobos group, serum CRP levels in high, medium and low concentration HPS groups were significantly reduced (P <0.05); The results of pathological section: Compared with Dobasia group, the rats in HPS high-dose group had the most obvious retinal thickening, and the cells in each layer were arranged neatly. Compared with the control group, the levels of VEGF and PEDF in the retina were significantly increased in the blank group (P <0.05). Compared with the blank group, VEGF and PEDF showed that VEGF Level decreased, PEDF content increased, the difference was statistically significant (P <0.05). Conclusion: Hedysari polysaccharide can reduce the content of CRP in serum, decrease the level of VEGF in retinal tissue, increase the content of PEDF, restrain the production of retinal new blood vessels and protect the retina.
