粳型水稻Y98149是从离子束诱变后代中获得的显性半矮秆突变体。本研究用RAPD技术对突变体和野生型进行了DNA指纹分析,经506个10bp的随机引物的扩增,获得4个多态性差异片段,可用作显性半矮秆基因的分子标记。RAPD的结果也表明半矮秆突变体与野生型之间基因组差异十分微小,为突变基因的鉴定提供了分子生物学证据。
Japonica rice Y98149 is a dominant semi-dwarf mutant obtained from the offspring of ion beam mutants. In this study, DNA fingerprinting of mutants and wild-type was carried out by RAPD technique. Four fragments with different polymorphisms were obtained by amplification of 506 bp random primers, which could be used as molecular markers of dominant semi-dwarfing genes. The results of RAPD also showed that the genome difference between semi-dwarf mutant and wild type was very small, which provided the molecular biological evidence for the identification of the mutant gene.
