羰基镍急性中毒大鼠血清镍变化与淋巴细胞DNA损伤及加合物形成的研究

目的观察羰基镍急性中毒大鼠血清镍变化与淋巴细胞DNA损伤及加合物形成情况,探讨羰基镍急性中毒致大鼠淋巴细胞DNA损伤的机制。方法染毒后第1、2、3和7天,测定4个染毒组(A、B、C组分别为低、中、高浓度羰基镍染毒组,D组为氯气染毒组)与正常对照组(E组)大鼠的血清镍。采用紫外吸收光谱移动法测定淋巴细胞DNA加合物,单细胞凝胶电泳观察DNA损伤。结果各染毒组血清镍在染毒后第1天最高,随时间的推移逐渐降低。各染毒组淋巴细胞DNA均在259 nm处有最高吸收峰,与E组(在260 nm处有最高吸收峰)比较位移均为+1 nm。A组淋巴细胞DNA最大吸光度在第3天降至最低点,B组在第7天降至最低点,C组和D组在第2天降至最低点,与E组比较,差异均有统计学意义(P<0.05)。各染毒组淋巴细胞DNA彗星尾长和Olive尾矩均大于正常对照组(P<0.05)。结论羰基镍急性染毒后第1天大鼠血清镍最高。染毒后第2天淋巴细胞DNA加合物形成最多,第3天淋巴细胞DNA损伤最重,第7天有所恢复。 Objective To investigate the changes of serum nickel and the DNA damage of lymphocytes and the formation of adducts in rats with acute carbonyl nickel poisoning, and to explore the mechanism of DNA damage in rat lymphocytes induced by nickel carbonyl nickel poisoning. Methods On the 1st, 2nd, 3rd and 7th days after exposure, four exposure groups (group A, B and C were low, medium and high concentration carbonyl nickel exposure groups and D exposure was chlorine exposure groups) and Serum nickel in normal control group (group E) rats. The DNA adducts of lymphocytes were determined by UV absorption spectroscopy and the DNA damage was observed by single cell gel electrophoresis. Results Serum nickel in each exposure group was the highest on the first day after exposure and gradually decreased with the passage of time. The DNA of lymphocyte in each group had the highest absorption peak at 259 nm, and the displacement was +1 nm compared with E group (the highest absorption peak at 260 nm). The maximal absorbance of DNA of lymphocytes in group A decreased to the lowest point on day 3, the lowest in group B on day 7, the lowest on day 2 in group C and group D, the difference was statistically significant compared with group E Significance (P <0.05). The DNA tail length and Olive tail moment of DNA of all the infected groups were higher than that of the normal control group (P <0.05). Conclusion Nickel carbonyl nickel was the highest one day after acute exposure to nickel. Lymphocyte DNA adduct formed the most on the 2nd day after the infection, the DNA damage of the lymphocyte was heaviest on the 3rd day, and recovered on the 7th day.
