目的观察乳腺癌组织中Shh基因表达变化并探讨其与术后复发的关系。方法留取152例经手术治疗的乳腺癌患者肿瘤及癌旁组织,采用实时荧光定量PCR技术检测乳腺癌及癌旁组织标本中Shh基因表达,患者随访截止日期为2015年2月28日,以乳腺癌组织中Shh mRNA相对表达量的中位值2.47为界值,将患者分为高表达组89例和低表达组63例,采用Kaplan-Meier生存分析法分析患者术后无复发生存时间。结果乳腺癌、癌旁组织中Shh mRNA相对表达量分别为2.48±0.43、1.25±0.29,两者比较,P<0.05;乳腺癌组织中Shh mRNA相对表达量与年龄和人类表皮生长因子受体表达有关(P均<0.05),年龄越小、人类表皮生长因子受体表达阳性,Shh mRNA相对表达量越高;非复发组、复发组乳腺癌组织中Shh mRNA相对表达量分别为1.98±0.42、2.87±0.47,两组比较,P<0.05;高表达组、低表达组术后复发率分别为23.6%、12.7%,两组比较,P<0.05;Kaplan-Meier生存分析显示,高表达组、低表达组患者中位无复发生存时间分别为29、59个月,两组比较,P<0.05。结论 Shh在乳腺癌组织中呈高表达,且与术后复发有关,Shh高表达可显著缩短患者术后无复发生存时间。
Objective To observe the changes of Shh gene expression in breast cancer and explore its relationship with postoperative recurrence. Methods The tumor and paracancerous tissues of 152 breast cancer patients undergoing surgery were enrolled. The expression of Shh gene was detected by real-time fluorescence quantitative PCR in breast cancer and its adjacent tissues. The follow-up date of patients was February 28, 2015. The median value of Shh mRNA relative expression in breast cancer was 2.47. The patients were divided into high expression group 89 cases and low expression group 63 cases. Kaplan-Meier survival analysis was used to analyze the postoperative recurrence-free survival time. Results The relative expression levels of Shh mRNA in breast cancer and adjacent normal tissues were 2.48±0.43 and 1.25±0.29, respectively. There was a significant difference between the two groups (P<0.05). The relative expression of Shh mRNA was correlated with age and human epidermal growth factor receptor expression in breast cancer tissues. The relative expression of Shh mRNA was higher in younger people and positive human epidermal growth factor receptor expression (P<0.05). The relative expression of Shh mRNA in non-recurrent and recurrent breast cancer tissues was 1.98±0.42, respectively. 2.87±0.47, P<0.05 for the two groups; the recurrence rates for the high-expression group and the low-expression group were 23.6% and 12.7%, respectively; P<0.05 for the two groups; Kaplan-Meier survival analysis showed that the high expression group, The median time to recurrence-free survival in the low-expression group was 29 and 59 months, respectively, with a P<0.05 difference between the two groups. Conclusion Shh is highly expressed in breast cancer tissues and is associated with postoperative recurrence. Shh high expression can significantly reduce the postoperative recurrence-free survival time.
