PPARγ天然激动剂姜黄素对自发性高血压大鼠颈动脉中膜组织学变化的影响

目的明确PPARγ天然激动剂姜黄素对自发性高血压大鼠颈动脉中膜组织学变化的影响,分析姜黄素对高血压动脉粥样硬化防治的应用前景。方法 12周龄雄性自发性高血压大鼠20只,随机分为姜黄素干预组(姜黄素200mg/kg.d灌胃给药处理)和模型组(无任何干预的自发性高血压大鼠组),同周龄雄性WKY大鼠10只作为正常血压对照组。大鼠尾动脉无创血压测定方法测定大鼠清醒安静状态下尾动脉的收缩压。喂养至18周龄时终止实验处死动物取颈动脉标本,检测平滑肌肌动蛋白,测定内外弹力膜间血管中膜的厚度、横截面积、平滑肌细胞核面积。结果平滑肌肌动蛋白在3组大鼠中膜中均大量表达;实验开始至实验结束所测的每个时间点均显示模型组大鼠及姜黄素干预组收缩压较WKY大鼠收缩压显著增高,随着鼠龄的增加,模型组及姜黄素干预组大鼠收缩压呈现逐渐增加的趋势,姜黄素干预组和模型组大鼠组在同一时间点收缩压比较无显著性差异;自发性高血压大鼠模型组及姜黄素干预组中膜厚度、中膜横截面积、平滑肌细胞核面积较WKY大鼠增加显著,姜黄素干预组上述3指标较高血压大鼠组明显下降。结论姜黄素作为PPARγ的天然激活剂,能够降低高血压大鼠颈动脉中膜的病理重构,提示姜黄素在动脉粥样硬化的防治中具有一定的应用前景。 Objective To investigate the effect of curcumin, a natural agonist of PPARγ, on the carotid intima-media histopathological changes in spontaneously hypertensive rats, and to analyze the application of curcumin in the prevention and treatment of hypertension-induced atherosclerosis. Methods Twenty male spontaneous hypertensive rats at 12 weeks of age were randomly divided into four groups: curcumin intervention group (200mg / kg.d) and model group (spontaneously hypertensive rats without any intervention group ), 10 male WKY rats of the same age as normal blood pressure control group. Measurement of rat tail artery noninvasive blood pressure measurement of the rat spermatic resting tail artery systolic pressure. Animals were sacrificed at 18 weeks of age to sacrifice the carotid arteries and the smooth muscle actin was measured. The thickness, cross-sectional area and smooth muscle cell nuclear area of ​​the medial and lateral elastic membranes were measured. Results Smooth muscle actin was highly expressed in the media of all three groups. At each time point from the start of the experiment to the end of the experiment, the systolic blood pressure of model group and curcumin intervention group was significantly higher than that of WKY group , Systolic blood pressure of model group and curcumin intervention group increased gradually with the increase of mice’s age, and there was no significant difference in systolic blood pressure between curcumin intervention group and model group at the same time point; spontaneous high Blood pressure rat model group and curcumin intervention group medial thickness, medial membrane cross-sectional area, smooth muscle cell nuclear area increased significantly than WKY rats, curcumin intervention group of the above three indicators of hypertension decreased significantly. Conclusion As a natural activator of PPARγ, curcumin can reduce the pathological remodeling of carotid artery in hypertensive rats, suggesting that curcumin may have some application prospects in the prevention and treatment of atherosclerosis.
