目的研究香烟对肺部炎性介质IL-1I、L-6I、L-8分泌的影响。方法制备香烟烟雾提取物(CSE),用不同浓度的CSE(10%~100%)刺激肺泡Ⅱ型上皮细胞A549,以放射免疫法检测细胞培养上清液中IL-1I、L-6I、l-8的水平。用不同吸烟剂量(低、中、高剂量组分别每天吸烟4、8、16支)制备动物吸烟模型,采用自主开发的吸烟机给烟12周,以放免法检测肺灌洗液(BALF)中IL-1I、L-6I、L-8的水平。结果各浓度CSE组IL-1水平与对照组相比,差异无统计学意义(P>0.05),20%、50%、100%CSE组IL-6的水平明显升高I,L-8的水平下降,差异有统计学意义(P<0.05);各吸烟剂量组IL-1的水平与对照组相比,差异无统计学意义(P>0.05),低、中、高剂量组IL-6I、L-8的水平显著升高,差异有统计学意义(P<0.05)。结论香烟会影响肺部炎性介质IL-6I、L-8的分泌。
Objective To study the effects of cigarette smoking on IL-1I, L-6I and L-8 secretion in inflammatory mediators of the lungs. Methods Cigarette smoke extract (CSE) was prepared. A549 cells were stimulated with different concentrations of CSE (10% -100%) and IL-1I, L-6I, -8 level. Animals were smoked at different smoking doses (4, 8, and 16 cigarettes per day for low, middle and high dose groups) respectively. Tobacco was smoked for 12 weeks in a self-developed smoking machine and BAL was measured by radioimmunoassay IL-1I, L-6I, L-8 levels. Results The levels of IL-1 in CSE group were not significantly different from those in control group (P> 0.05). The levels of IL-6 in CSE group were significantly higher than those in control group (P <0.05). The levels of IL-1 in each smoking dose group were not significantly different from those in the control group (P> 0.05). IL-6I , L-8 levels were significantly increased, the difference was statistically significant (P <0.05). Conclusion Cigarettes can affect the secretion of inflammatory mediators IL-6I and L-8 in lung.
