目的 探讨 NF-κB在脂肪酸合酶抑制剂 ( FASI)诱导人结肠癌 Lo Vo细胞凋亡中的作用。方法 FASI—浅蓝菌素处理大肠癌 Lo Vo细胞株 ,四唑盐比色试验 ( MTT法 )、片断 DNA琼脂糖凝胶电泳、流式细胞仪及荧光染色 ( AO/EB)等方法检测细胞调亡 ,用凝胶电泳迁移率法 ( EM-SA)检测 NF- κB活性。结果 Lo Vo细胞在 FASI作用下 ,细胞增殖被阻遏 ,并呈现剂量效应关系。同时诱导细胞发生凋亡。FASI对人成纤维细胞增殖无明显影响。NF- κB在不同浓度浅蓝菌素 ( 1 0 -9~ 1 0 -5)处理 Lo Vo细胞 1 2 h后 ,可抑制 NF- κB活性 ,其抑制程度呈剂量依赖性递减。结论FASI可能通过抑制 Lo Vo细胞内源性脂肪酸合成 ,诱发细胞凋亡来阻遏 Lo Vo的增殖。NF- κB可能参与 FASI诱导人结肠癌 Lo Vo细胞凋亡中的信号转导
Objective To investigate the role of NF-κB in fatty acid synthase inhibitor (FASI)-induced apoptosis of human colon cancer Lo Vo cells. Methods FASI-cerulenin treatment of colon cancer Lo Vo cells, tetrazolium salt colorimetric assay (MTT method), fragment DNA agarose gel electrophoresis, flow cytometry and fluorescence staining (AO/EB) and other methods to detect cells After apoptosis, NF-κB activity was measured by gel electrophoresis mobility shift assay (EM-SA). RESULTS: Lo Vo cells were inhibited by FASI and the proliferation of cells was dose-dependent. At the same time induce apoptosis of the cells. FASI has no significant effect on the proliferation of human fibroblasts. NF-κB inhibited the activity of NF-κB in Lo Vo cells after treatment with different concentrations of cerulenin ( 1 0 -9 - 1 0 -5) for 12 hours, and its inhibition was dose-dependently decreased. Conclusion FASI may inhibit the proliferation of Lo Vo by inhibiting the synthesis of endogenous fatty acids in Lo Vo cells and inducing apoptosis. NF-κB may be involved in FASI-induced signal transduction in human Colon cancer Lo Vo cells
