目的了解舟山渔场海洋鱼类异尖线虫感染状况。方法从舟山市农贸市场购买带鱼、小黄鱼和鲐鱼3个品种89尾鱼,解剖后在光学显微镜下观察异尖线虫幼虫形态并进行初步鉴定;采用限制性片段长度多态性聚合酶链反应(PCR-RFLP)方法分析核糖体DNA(r DNA)内转录间隔区(ITS)进行分子学鉴定。结果在82尾鱼中检出异尖线虫幼虫,感染率为92.13%,共获得1 023条异尖线虫样品。鱼小肠、肠系膜和性腺(62.35%)以及体腔周围脏器(37.05%)是主要的寄生部位。随机选择435条异尖线虫进行PCR-RFLP分子鉴定,结果显示有异尖属和宫脂属2个属的7种异尖线虫,包括简单异尖线虫(A.simplex s.s.,占4.60%)、派氏异尖线虫(A.pegreffii,占81.61%)、典型异尖线虫(A.typica,占0.23%)、内弯宫脂线虫(H.aduncum,占2.07%)、费氏宫脂线虫(H.fabri,占3.91%)、宫脂线虫未定种(H.spp,占2.99%)和厦门宫脂线虫(H.amoyense 4.37%)。结论舟山渔场带鱼、小黄鱼和鲐鱼异尖线虫感染情况较为严重,派氏异尖线虫为感染优势种。
Objective To understand the infection of Anisakis in marine fish in Zhoushan Fishing Ground. Methods Three hundred and eight species of fish, octopus, small yellow croaker and bighead carp were purchased from Zhoushan Farmer’s Market. The morphology of the larvae was observed under light microscope after the dissection. The restriction fragment length polymorphism (PCR) -RFLP) was used to analyze the molecular structure of the internal transcribed spacer (ITS) in ribosomal DNA (rDNA). Results Anisakis larvae were detected in 82 fish, and the infection rate was 92.13%. A total of 1 023 Anaxis nematodes samples were obtained. The small intestine, mesentery and gonads (62.35%) and the organs around the body cavity (37.05%) were the main parasitic sites. A total of 435 Achilles’ nematodes were randomly selected for PCR-RFLP molecular analysis. The results showed that there were 7 kinds of ANA, including A.simplex ss (4.60%), A. pegreffii (81.61%), typical A.typica (0.23%), H. aduncum (2.07%), C. adipose H.fabri, accounting for 3.91%), Uncaria tomentosa (H.spp, accounting for 2.99%), and H.amoyense (4.37%). Conclusion The infection of Anisakis ocellatus in small fish and yellow croaker in Zhoushan fishing ground is more serious. Anisakis vicinae is the dominant species of infection.
