目的建立以高效液相色谱法测定人尿中酮洛酸浓度的方法。方法色谱柱为Waters Sunfire C18,流动相为0.02mol·L-1磷酸二氢钾溶液-乙腈(70∶30,用磷酸调节pH值至4.5),检测波长为315nm,柱温为35℃,流速为1mL.min-1,进样量为20μL,保留时间为9.1min。结果人尿中酮洛酸浓度在0.1~10.0μg·mL-1范围内线性关系良好,日内和日间RSD<10%,最低定量限为0.1μg·mL-1。结论本法简便、快捷、灵敏,适用于酮洛酸人体药动学研究。
Objective To establish a method for the determination of ketorolac in human urine by high performance liquid chromatography. Methods The column was Waters Sunfire C18. The mobile phase consisted of 0.02 mol·L-1 potassium dihydrogen phosphate solution - acetonitrile (70:30, adjusted to pH 4.5 with phosphoric acid). The detection wavelength was 315 nm, the column temperature was 35 ℃, the flow rate 1mL.min-1, injection volume of 20μL, retention time of 9.1min. Results The concentration of ketorolac in human urine had a good linearity in the range of 0.1-10.0 μg · mL-1 with a RSD of <10% at intra-day and inter-day and a minimum limit of quantification of 0.1 μg · mL-1. Conclusion This method is simple, rapid and sensitive, suitable for the pharmacokinetics of ketorolac.
