利用扩增片段长度多态性(Amplified fragment length polymorphism,AFLP)分子标记技术,对26份鸡冠花种质资源进行亲缘关系探讨。结果表明:1)采用8对引物组合共扩增出8 063条谱带,其中多态性条带为7 959条,多态性条带比率为98.71%,平均每对引物扩增出1 007.9个位点和994.9个多态性位点。26份种质的相似性系数居于0.264 2~0.664 8,平均值为0.464 5,每个位点的有效等位基因数为1.078 0,平均多样性指数为0.208 7,遗传相似性系数最高的是1和3号,达到0.664 8,最低的是13和24号,为0.264 2。根据遗传相似性系数进行聚类分析,在遗传相似性系数0.46处切割时,可将图中26个鸡冠花种质资源分为3个类群:24号杂交种粉色头状鸡冠花单独聚为一类,8号多头红色鸡冠花单独聚为一类;其他24个鸡冠花材料聚为一类。根据不同的花型和花色,24个鸡冠花材料又可分成6个小类群。分子聚类结果和形态聚类结果较为一致。24号杂交粉色头状鸡冠花和8号多头鸡冠花‘威信’,与其他24个鸡冠花材料的亲缘关系较远;来源于相同地区的种质资源,如19和25号同为来自于甘肃的品种亲缘关系较近;来自科研所自育品种16和23号单独聚为一类,与其他种质的亲缘关系较远。
The genetic relationship of 26 cockroach germplasm resources was explored by using AFLP molecular marker technique. The results showed as follows: 1) A total of 8 063 bands were amplified by 8 pairs of primers, of which 7 959 bands were polymorphic bands and 98.71% bands were polymorphic bands, with an average of 1 007.9 Sites and 994.9 polymorphic sites. The similarity coefficient of 26 germplasms ranged from 0.264 2 to 0.664 8 with an average of 0.464 5. The number of effective alleles at each locus was 1.078 0 and the average diversity index was 0.208 7. The highest genetic similarity coefficient was 1 and 3, reaching 0.664 8, the lowest being 13 and 24, 0.264 2. According to the genetic similarity coefficient, cluster analysis was conducted. When the genetic similarity coefficient was 0.46, the 26 germplasm resources were divided into three groups: Class, No. 8 long red celosia grouped together into one category; the other 24 celosia materials clustered into one category. According to different patterns and colors, 24 celosia materials can be divided into 6 sub-groups. Molecular clustering results and morphological clustering results are more consistent. The hybrid pink head-shaped cockscomb flower No.24 and the long-handled cockscomb No.8, which is the long-handled hybrid flower of Cixi, were distantly related to the other 24 cockscomb materials. Germplasm resources from the same area, such as 19 and 25, Of the species closer genetic relationship; from the Institute of self-breeding varieties 16 and 23 together as a separate category, distant genetic relationship with other germplasm.
