目的:研究Fhit基因在鼻腔鼻窦恶性肿瘤中的表达及临床意义,探讨其相关性。方法:采用免疫组织化学SP法检测48例原发性鼻腔鼻窦恶性肿瘤组织、29例癌旁正常组织及21例良性病变组织中Fhit基因的表达;采用碘化丙啶(PI)及苏木精伊红染色检测肿瘤细胞自发性凋亡情况。结果:48例中,26例(54.17%)Fhit表达减少或缺失,22例(45.83%)表达正常。Fhit表达的平均A值与凋亡指数(AI)成正相关(r=0.379,P<0.01)。Fhit表达正常者预后显著好于表达减少或缺失者(P<0.05)。结论:Fhit基因失活与鼻腔鼻窦恶性肿瘤的发生、预后相关,可能是该肿瘤的抑癌基因,其抑癌作用可能通过促进细胞凋亡实现,Fhit基因表达减弱有可能成为鼻腔鼻窦恶性肿瘤新的预后指标。
Objective: To study the expression and clinical significance of Fhit gene in nasal sinus cancer and to explore its correlation. Methods: Immunohistochemical SP method was used to detect the expression of Fhit gene in 48 cases of primary nasal sinus malignant tumor tissues, 29 cases of adjacent normal tissues and 21 cases of benign lesions. Propofol (PI) and hematoxylin Eosin staining detection of spontaneous apoptosis of tumor cells. Results: In 48 cases, Fhit expression was decreased or disappeared in 26 cases (54.17%), and normal expression was found in 22 cases (45.83%). The average A value of Fhit expression was positively correlated with apoptosis index (r = 0.379, P <0.01). Fhit normal expression was significantly better than those with reduced expression or deletion (P <0.05). CONCLUSION: Inactivation of Fhit gene is associated with the occurrence and prognosis of nasal sinus malignant tumors, and may be the tumor suppressor gene of this tumor. The anti-tumor effect of Fhit may be achieved through the promotion of apoptosis. Fhit gene expression may be a new malignant tumor of nasal sinus The prognostic indicators.
