利用不同大豆品种的顶芽、胚轴、子叶节、子叶、叶片等做为外植体,进行组织培养再生研究。结果表明:不同大豆品种诱导分化芽的能力差别较大,供试品种中,辽豆11号和辽豆10号的再生能力较强,诱芽率分别为39.8%和36.0%。对诱芽率的影响,经L_(16)(4)~5正交实验可知外植体>品种>培养基。外值体对诱芽率影响依次为:子叶节>顶芽>子叶>下胚轴。6-BA对不同外植体诱导芽发生的适宜浓度不同,顶芽、下胚轴为0.5mg·l~(-1),子叶为1.0mg·1~(-1),成熟子叶为4.0mg·L~(-1)。
The top buds, hypocotyls, cotyledons, cotyledons and leaves of different soybean cultivars were used as explants to study the regeneration of tissue culture. The results showed that the ability of different soybean varieties to differentiate buds was quite different. Among the tested varieties, the regeneration ability of Liaodou 11 and Liaodou 10 were stronger with the rate of 39.8% and 36.0% respectively. On the rate of budding, explants> variety> medium were found by L_ (16) (4) ~ 5 orthogonal experiment. The effect of outer body on the rate of seed germination were as follows: cotyledonary node> terminal bud> cotyledon> hypocotyl. The optimum concentrations of 6-BA for inducing buds of different explants were different. The top bud, hypocotyls were 0.5 mg · L -1, the cotyledons were 1.0 mg · 1 -1, and the mature cotyledons were 4.0 mg · L -1.
