三种对证不同高血压证型的中药复方对SHR影响的蛋白组学比较分析

目的:考察治疗不同证型高血压的半夏白术天麻汤、地黄饮子和天麻钩藤饮对自发性高血压大鼠肾脏蛋白谱表达差异,并通过主成分分析综合比较,阐述三种复方的作用特点与调控通路,进而分析高血压分型治疗的理论基础。方法:6周龄的自发性高血压大鼠(Spontaneous hypertensive rat,SHR)与正常对照大鼠(Wistar-Kyoto rat,WKY),对给药组的SHR分别灌胃天麻钩藤饮、半夏白术天麻汤和地黄饮子3种复方,监测鼠尾血压变化至24周龄,统一处死提取肾脏组织蛋白并进行双向电泳实验,经PDQuest软件分析得差异蛋白点,结合肾脏蛋白表达谱以主成分分析法(Principal component analysis,PCA)考察三种复方对SHR的作用特点。结果:SHR从6周龄至10周龄血压处于持续升高状态,此时天麻钩藤饮具有良好的降压效果;当血压稳定保持较高水平时,地黄饮子与半夏白术天麻汤的降压效果优于天麻钩藤饮;24周龄时肾脏蛋白谱分析共找出SHR与WKY表达显著不同的84个蛋白点,结合主成分分析,累计主成分提取95.951%,发现天麻钩藤饮和半夏白术天麻汤对于SHR肾脏蛋白点的改变有显著变化趋势,且向WKY趋近,地黄饮子的作用趋势较为分散。结论:在SHR血压升高初期,天麻钩藤饮具有显著的降压效果;当血压稳定保持较高水平,地黄饮子与半夏白术天麻汤的降压效果优于天麻钩藤饮。半夏白术天麻汤和天麻钩藤饮对SHR肾脏蛋白表达的改善有向WKY回归的趋势,地黄饮子作用较为分散。 OBJECTIVE: To investigate the difference of kidney protein expression of Banxiabaizhu Tianma Decoction, Dihuang Yinzi and Tianma Gouteng Decoction in spontaneously hypertensive rats in the treatment of hypertension with different syndromes. Through the comparision of principal component analysis and comprehensive comparison, the effects of three compounds Role characteristics and regulatory pathway, and then analyze the theoretical basis of hypertension treatment. Methods: Six weeks old spontaneous hypertensive rats (SHR) and normal control rats (Wistar-Kyoto rat, WKY) were administered with SHR, Tianma Decoction and Rehmanniae Decoction three kinds of compound to monitor the changes in rat tail blood pressure to 24 weeks of age, unified sacrificial renal tissue protein extraction and two-dimensional electrophoresis experiments, the PDQuest software analysis of the differential protein spots, combined with kidney protein expression profile of the principal component analysis (Principal component analysis, PCA) to investigate the effect of three kinds of compound on SHR. Results: From 6 weeks to 10 weeks of age, the blood pressure of SHR was continuously elevated. At this time, Tianma Gouteng Decoction had a good antihypertensive effect. When blood pressure remained high, Dihuang Yinzi and Tianxia Decoction Antihypertensive effect was better than that of Tianma Gouteng Decoction. At 24 weeks of age, 84 protein spots with significantly different SHR and WKY expression were found, and 95.951% of the total principal components were extracted by principal component analysis. And Pinelliae Tianma Decoction for SHR kidney protein point changes have a significant trend, and approached WKY, Rehmannia decoction of the role of the trend is more dispersed. CONCLUSION: Tianma Gouteng Decoction has a significant antihypertensive effect in the early period of SHR blood pressure. When the blood pressure is kept at a high level, the antihypertensive efficacy of Dihuang Yinzi and Banxia Baizhu Tianma Decoction is better than that of Tianma Gouteng Decoction. Pinellia Atractylodes Tianma and Tianma Gouteng Decoction on the improvement of protein expression in SHR WKY regression trend, Rehmannia decoction more dispersed.
