目的 测定根据计算机预测的SARS冠状病毒S蛋白S1区的抗原表位多肽与SARS病人血清的免疫反应,以寻找和确定SARS相关冠状病毒的中和抗原表位。方法 采用多肽合成仪合成SARS冠状病毒S蛋白S1区的多肽,并用ELISA方法检测合成的多肽与SARS病人血清的免疫反应。结果 合成的8个多肽与SARS病人血清免疫反应的光密度值是其与正常人血清免疫反应的光密度值的1.5~2.6倍。结论 合成的多肽与SARS病人血清的免疫反应较低,提示这些多肽是否为SARS冠状病毒的抗原表位还要进一步研究。
OBJECTIVE: To determine the immunological reaction of SARS-CoV S protein S1 epitope polypeptide and SARS patient serum in order to find out and identify the SARS-associated coronavirus neutralizing epitopes. Methods Peptide of S1 protein of SARS coronavirus S protein was synthesized by peptide synthesizer and the immune responses of the synthesized peptides and serum of SARS patients were detected by ELISA. Results The optical density of the immunogenic reaction of the eight peptides synthesized with SARS was 1.5 ~ 2.6 times higher than that of the normal human serum. Conclusions The immunogenic reaction of the synthesized peptides with the serum of SARS patients is low, suggesting that whether these polypeptides are epitopes of the SARS-CoV should be further studied.
