目的:研究虫草素对A549细胞增殖、凋亡及核转录因子NF-κB的影响,并探讨其诱导凋亡的机制。方法:采用MTS法检测不同浓度虫草素对A549细胞的增殖抑制作用,并计算24 h的IC50;用Hoechst 33258染色法及流式细胞术检测24 h不同浓度虫草素诱导A549细胞凋亡的情况;Western blot法检测虫草素对A549细胞核内NF-κB p65及凋亡相关蛋白BAX、BCL-2、cleaved Caspase-3表达的影响。结果:虫草素能明显抑制A549细胞增殖,促进其凋亡;随着虫草素浓度的升高,BAX和cleaved Caspase-3的表达上调,而BCL-2表达下调;Western blot检测结果显示,虫草素能抑制A549细胞质中的NF-κB p65向核内转位。以上作用均呈现量效关系。结论:虫草素能抑制A549细胞增殖并促进其凋亡,机制可能与抑制NF-κB通路相关。
Objective: To study the effects of cordycepin on the proliferation and apoptosis of A549 cells and the nuclear transcription factor NF-κB, and to explore its mechanism of apoptosis. Methods: The inhibitory effects of cordycepin at different concentrations on the proliferation of A549 cells were detected by MTS method. The IC50 of 24 h was calculated. Hoechst 33258 staining and flow cytometry were used to detect the apoptosis of A549 cells induced by different concentrations of cordycepin. The effect of cordycepin on the expression of NF-κB p65 and apoptosis related proteins BAX, BCL-2 and cleaved Caspase-3 in A549 cells was detected by Western blot. Results: Cordycepin significantly inhibited the proliferation of A549 cells and promoted its apoptosis. With the increase of cordycepin concentration, the expression of BAX and cleaved Caspase-3 was up-regulated and the expression of BCL-2 was down-regulated. Western blot showed that cordycepin Can inhibit the nuclear translocation of NF-κB p65 in the A549 cytoplasm. The above effects show a dose-effect relationship. Conclusion: Cordycepin can inhibit A549 cell proliferation and promote its apoptosis, which may be related to the inhibition of NF-κB pathway.
