目的探讨葡萄籽提取物原花青素(grape seed procyanidin extract,GSPE)对人膀胱癌BIU87细胞周期的影响及其可能机制。方法应用不同浓度GSPE(50、100、200μg/ml)与膀胱癌BIU87细胞共同培养24 h后,采用流式细胞术检测细胞周期,RT-PCR、Western blot检测CyclinD1、CDK4mRNA及蛋白表达变化。结果GSPE可呈剂量依赖性阻滞细胞于G1期,并可引起CyclinD1和CDK4mRNA及蛋白水平的表达减少(P<0.01)。结论GSPE可诱导人膀胱癌BIU87细胞周期阻滞,其机制可能是通过下调CyclinD1和CDK4的表达而实现。
Objective To investigate the effect of grape seed extract procyanidins extract (GSPE) on the cell cycle of human bladder cancer BIU87 and its possible mechanism. Methods Cell cycle was detected by flow cytometry after co-cultured with different concentrations of GSPE (50, 100, 200 μg/ml) and bladder cancer BIU87 cells for 24 h. The expression of CyclinD1 and CDK4 mRNA and protein were detected by RT-PCR and Western blot. RESULTS: GSPE inhibited the G1 phase in a dose-dependent manner, and decreased the expression of CyclinD1 and CDK4 mRNA and protein (P<0.01). Conclusion GSPE can induce cell cycle arrest in human bladder cancer BIU87. The mechanism may be achieved by down-regulating the expression of CyclinD1 and CDK4.
