目的 了解异氟醚对大鼠脑 3’5’ -环腺苷酸 (cAMP)含量的影响。方法 SD大鼠 40只 ,随机分为五组 ,分别在未吸入异氟醚 (对照组 ) ,吸入 1.4%异氟醚翻正反射消失时 (翻正反射消失组 ) ,吸入 1.4%异氟醚平衡 30min时 (麻醉组 ) ,吸入 1.4%异氟醚平衡 30min后将动物取出自制麻醉箱待翻正反射恢复时 (恢复Ⅰ组 )和吸入 1.4%异氟醚平衡 30min后将动物取出自制麻醉箱后 30min时 (恢复Ⅱ组 )断头取脑。应用蛋白竞争结合分析法测定大脑皮层、脑干的cAMP含量。结果 与对照组比较 ,仅麻醉组大脑皮层cAMP含量明显升高 ,是对照组的 149% (P <0 .0 1) ,翻正反射消失组、麻醉组及恢复Ⅰ组的脑干cAMP含量分别是对照组的 137%、177%和 12 8% ,均明显升高 (P <0 0 1)。结论 吸入异氟醚时大脑皮层、脑干AMP含量的升高 ,在异氟醚的全麻分子学机制中可能发挥重要作用。
Objective To investigate the effect of isoflurane on 3’5 ’- cAMP content in rat brain. Methods Forty Sprague-Dawley rats were randomly divided into five groups, respectively. When isoflurane was not inhaled (control group), inhaled 1.4% isoflurane norbital reflex disappeared Animals were taken out of homemade anesthesia box after 30 minutes of equilibration (anesthesia group), 1.4% isoflurane imbalanced 30 minutes after inhalation, and animals were taken out of homemade anesthesia box After 30min (recovery group Ⅱ) decapitated brain. The cAMP content in cerebral cortex and brainstem was determined by protein-binding assay. Results Compared with the control group, the cAMP content in the cerebral cortex of the anesthetized group was significantly higher than that of the control group (149%, P <0.01). The levels of cAMP in the brainstem of the normalized reflex group, the anesthetic group and the recovery group were Which is 137%, 177% and 12 8% of the control group, both significantly increased (P <0.01). Conclusion The increase of AMP content in cerebral cortex and brainstem during inhalation of isoflurane may play an important role in the molecular mechanism of general anesthesia of isoflurane.
