目的:探讨大鼠睾丸组织COXⅦa(cytochrome C oxidase Ⅶ a)表达的加龄变化及其与睾酮合成之间的相关性。方法:①选用出生后1周、3周、5周、3月和24月龄雄性SD大鼠,每组6只。②采用半定量RT-PCR法检测大鼠睾丸组织COXⅦamRNA的表达。③Western blotting方法检测大鼠睾丸组织COXⅦa蛋白表达。④化学发光法检测大鼠血清睾酮浓度。结果:①血清睾酮浓度3月、24月、5周、3周和1周龄组依次降低,其中3月龄和24月龄组与其余各组间差异有统计学意义(P<0.05)。②COXⅦamRNA水平24月、1周、3周、5周和3月龄组依次降低,其中3月龄和24月龄组与其余各组之间差异有统计学意义(P<0.05)。③睾丸组织COXⅦa蛋白表达,24月龄组最强,3月龄组最弱,除1周龄和3周龄组之间差异无统计学意义外,其余各组之间差异均有统计学意义(P<0.05)。④睾酮浓度与COXⅦamRNA水平和蛋白表达呈负相关(r=-0.580,P=0.013;r=-0.447,P=0.000),而COXⅦamRNA水平和蛋白表达呈正相关(r=0.941,P=0.000)。结论:COXⅦa随增龄变化,老年大鼠COXⅦa显著上调,与睾酮合成能力下降有关,可能成为治疗PADAM的候选基因。
Objective: To investigate the age-related changes in the expression of COXⅦa and its correlation with testosterone synthesis in rat testes. Methods: ① Male Sprague-Dawley rats, 1 week, 3 weeks, 5 weeks, 3 months and 24 months after birth, were selected. ② The semi-quantitative RT-PCR was used to detect the expression of COXⅦamRNA in rat testis. ③ Western blotting was used to detect the COXⅦa protein expression in rat testis. ④ chemiluminescence detection of serum testosterone in rats. Results: ① Serum testosterone levels decreased in March, 24, 5, 3 and 1 week groups, with statistical significance (P <0.05) between the 3 and 24 month old groups. (2) COXⅦamRNA levels decreased sequentially in 24 months, 1 week, 3 weeks, 5 weeks and 3 months, among which the difference was statistically significant (P <0.05) between 3 months and 24 months group and the other groups. ③ The expression of COXⅦa protein in testis tissue was the strongest in 24-month-old group and the weakest in 3-month-old group, with no significant difference except 1-week-old group and 3-week-old group (P <0.05). ④There was a negative correlation between testosterone concentration and COXⅦamRNA level and protein expression (r = -0.580, P = 0.013; r = -0.447, P = 0.000), while there was a positive correlation between COXⅦamRNA level and protein expression (r = 0.941, P = 0.000). CONCLUSION: The changes of COXⅦa in aged rats are significantly up-regulated, which may be related to the decreased ability of testosterone synthesis. It may be a candidate gene for the treatment of PADAM.
