目的研究阿司匹林对小鼠腹腔巨噬细胞基质金属蛋白酶(MMP)-2/9和基质金属蛋白酶组织抑制因子-1(TIMP-1)基因表达及分泌的影响。方法培养的小鼠腹腔巨噬细胞用阿司匹林处理后,用RT-PCR方法检测MMP-2/9和TIMP-1mRNA表达,用ELISA方法测定培养上清液中MMP-2/9和TIMP-1的含量。结果小鼠腹腔巨噬细胞经用阿司匹林12.5~50μg/ml处理24h后,MMP-2/9mRNA表达及分泌至培养上清液中的含量明显降低,MMP-2的降低百分率分别为18.6%~86.8%和37.7%~53.3%,MMP-9的降低百分率分别为32.1%~78.3%和6.5%~14.3%;而TIMP-1mRNA的表达及分泌却显著提高,其增加百分率分别为60.5%~110.9%和94.4%~80.6%。结论阿司匹林可抑制MMP-2/9的基因表达及分泌,同时还可增加TIMP-1的基因表达及分泌,这一效应对增加动脉粥样硬化斑块的稳定性和预防心血管事件的发生具有有利的作用。
Objective To study the effect of aspirin on the expression and secretion of matrix metalloproteinase (MMP) -2/9 and tissue inhibitor of metalloproteinase-1 (TIMP-1) in murine peritoneal macrophages. Methods The cultured mouse peritoneal macrophages were treated with aspirin, and the expressions of MMP-2/9 and TIMP-1 mRNA were detected by RT-PCR. The expressions of MMP-2/9 and TIMP-1 in culture supernatants content. Results After intraperitoneal injection of 12.5 ~ 50μg / ml aspirin for 24 hours, the expression of MMP-2/9 mRNA in the peritoneal macrophages and the secretion of MMP-2/9 mRNA in the culture supernatant decreased significantly. The percentages of MMP-2 decreased from 18.6% % And 37.7% -53.3% respectively. The percentages of MMP-9 decreased from 32.1% to 78.3% and from 6.5% to 14.3%, while the expression and secretion of TIMP-1 mRNA increased significantly from 60.5% to 110.9% And 94.4% ~ 80.6%. Conclusion Aspirin can inhibit the gene expression and secretion of MMP-2/9 and increase the gene expression and secretion of TIMP-1 at the same time. This effect has the effect of increasing the stability of atherosclerotic plaque and preventing the occurrence of cardiovascular events Beneficial effect.
