目的:探讨胃腺癌组织Notch1和Jagged1蛋白的表达及其与临床病理学特征的关系和意义。方法:采用免疫组织化学SP法检测2009-01-01-2012-12-12泰山医学院附属莱芜医院60例胃腺癌组织和癌旁组织中Notch1和Jagged1蛋白表达水平。结果:Notch1在胃腺癌组织中的阳性表达率为41.7%(25/60),癌旁组织为81.7%(49/60),差异有统计学意义,χ2=20.31,P<0.001;Jagged1在胃腺癌组织中的阳性表达率为71.7%(43/60),癌旁组织为96.7%(58/60),差异有统计学意义,χ2=14.07,P<0.001。Notch1表达与肿瘤分化程度(P=0.006)、淋巴结有无转移(P=0.005)和浸润深度(P=0.009)有关,Jagged1的表达与肿瘤分化程度(P=0.004)、浸润深度(P=0.009)和淋巴结有无转移(P=0.006)有关。Spearman相关分析结果显示,胃腺癌组织中Notch1和Jagged1蛋白表达呈正相关,r=0.460,P<0.001。结论:胃腺癌组织中Notch1和Jagged1蛋白表达下调,且两者呈正相关,可能与胃腺癌的发生、浸润及转移等多个环节有关;检测Notch1和Jagged1的表达有助于判断胃腺癌的生物学行为。
Objective: To investigate the expression of Notch1 and Jagged1 in gastric adenocarcinoma and its relationship with clinicopathological features. Methods: Immunohistochemical SP method was used to detect the expression of Notch1 and Jagged1 protein in 60 cases of gastric adenocarcinoma and paracancerous tissues at Laiwu Hospital affiliated to Taishan Medical College from January 2009 to January 2012. Results: The positive expression rates of Notch1 in gastric adenocarcinoma were 41.7% (25/60) and paracancerous tissues were 81.7% (49/60), the difference was statistically significant (χ2 = 20.31, P <0.001) The positive expression rates in cancer tissues were 71.7% (43/60) and 96.7% (58/60) in adjacent tissues, the difference was statistically significant (χ2 = 14.07, P <0.001). The expression of Notch1 was correlated with tumor differentiation (P = 0.006), lymph node metastasis (P = 0.005) and invasion depth (P = 0.009). The expression of Jagged1 was correlated with tumor differentiation ) And lymph node metastasis (P = 0.006). Spearman correlation analysis showed that the expression of Notch1 and Jagged1 in gastric adenocarcinoma was positively correlated, r = 0.460, P <0.001. Conclusion: The expressions of Notch1 and Jagged1 in gastric adenocarcinoma are down-regulated, and they are positively correlated with each other, which may be related to the occurrence, invasion and metastasis of gastric adenocarcinoma. Detecting the expression of Notch1 and Jagged1 may help to judge the biology of gastric adenocarcinoma behavior.
